摘要
为提高甜茶的综合利用价值,从提取甜茶甙废液中回收甜茶多酚并采用大孔树脂纯化的方法研究甜茶总多酚的纯化工艺条件;采用紫外分光光度法对甜茶多酚含量及其抗氧化活性进行测定。结果表明:所筛选的HPD826树脂为甜茶总多酚纯化的最佳树脂;当上样液的浓度为3.65mg/mL、上柱液体积为50mL、流速为1BV/h时,吸附率为94.25%,用30%的乙醇解吸后其甜茶总多酚的纯度可达56.68%;纯化过的甜茶总多酚对·OH和DPPH·自由基均有较好的清除作用,其IC50值分别为517.9μg/mL和45.9μg/mL。
In order to improve the comprehensive utilization of sweet tea,total polyphenol of sweet tea from which rubusoside liquid was extracted and the purification process conditions of total polyphenol from sweet tea were studied by macroporous resins purification method.The concentration of total polyphenol and its antioxidant activity was evaluated by ultraviolet spectrophotometry.The results showed that the resin of HPD826 was optimum for purification of total polyphenols from sweet tea,and when sample concentration was 3.65mg/mL,volume was 50 mL,sample flow rate was 1BV/h,adsorption rate was up to 94.25%,and the purity of total polyphenols was up to 56.68% by using 30% ethanol as eluent,purified the total polyphenol of sweet tea had better scavenging effect on · OH and DPPH ·,the determined IC50 values were 517.9μg/mL and 45.9μg/mL respectively.
