人参皂苷Rg_1、Rb_1对睡眠干扰所致SD大鼠认知和运动行为障碍的影响 被引量:9

Effects of Ginsenoside Rg_1 and Rb_1 on SD Rats' s Cognition and Exercise Behavior Disorder due to Sleep Interruption

摘要 目的:研究人参皂苷Rg1和Rb1对睡眠干扰致SD大鼠认知和运动行为障碍的影响。方法:将70只Sprague-Dawley雄性大鼠随机分为7组,每组10只,分别为:对照组(control)、模型组(model)、莫达非尼组(Modafinil,300 mg·kg-1)、Rg1高剂量组(Rg1,12 mg·kg-1)、Rg1低剂量组(Rg1,6 mg·kg-1)、Rb1高剂量组(Rb1,12 mg·kg-1)、Rb1低剂量组(Rb1,6 mg·kg-1)。滚筒适应3 d后开始连续15 d睡眠干扰造模并同时进行给药,于第15天开始自主活动实验、步态实验和Morris水迷宫实验。结果:经过连续15 d的睡眠干扰后,模型组大鼠的自主活动和步态行为均受到一定程度的下降,在水迷宫中潜伏期明显增加,穿台次数显著下降,而莫达非尼和人参皂苷Rg1和Rb1各给药组能不同程度地增加睡眠干扰大鼠的自主活动,增强大鼠左右肢的协调性,降低水迷宫中潜伏期。结论:两种人参皂苷Rg1和Rb1对睡眠干扰15 d后SD大鼠的认知和运动能力有改善作用。 Objective: To investigate the effects of ginsenoside RgI and Rbl on SD rats' s cognition and exercise behavior disorder due to sleep interruption. Methods: Seventy healthy adult male Sprague-Dawley rats were randomly divided into control group, model group, 300 mg·kg^-1 of Modafinil group, 4 treatment groups ( 6 mg·kg^-1, 12mg·kg^-1 of ginsenoside Rg1, 6 mg·kg^-1 , 12mg·kg^-1 of ginsenoside Rg1). After 3 days of adaptation, the rats were administrated orally the treatments for consecutive 15 days with sleep disturbances at the same time, and then adopted locomotor activity, gait instrument, and Morris water maze to elevate. Results: After 15 days of sleep disturbances, the locomotor activities and gait behaviors of the model group rats were reduced to a certain extent, the latency of place navigation in the Morris water maze was increased significantly and the number of crossing in the target quadranta was significantly reduced. While these indexes were improved to different extents after modafinil and ginsenosides Rg1 and Rb1 oral administration. Conclusion: Ginsenosides Rg1 and Rb1 have a significant improvement in cognitive and motor ability of SD rats after 15 days ' sleep interruption.
