神阙散治疗痛风性关节炎的药效学研究

Pharmacodynamic study of Shenque powder on gouty arthritis in rats

[目的]探讨神阙散对模型大鼠痛风性关节炎的治疗效果。[方法]分别建立大鼠高尿酸血症和痛风性关节炎模型,全自动生化分析测定大鼠的尿酸、肌酐、尿素氮,探讨神阙散对高尿酸血症的治疗作用;测定肿胀指数变化,同时取各组大鼠的骨组织进行石蜡包埋、苏木精-伊红（HE）染色进行病理学分析,评价神阙散对痛风性关节炎的治疗效果。[结果]两组实验组模型和对照均具有统计学差异,说明造模成功。对高尿酸血症,各给药组和模型对照组相比尿酸、肌酐、尿素氮均具有统计学意义,而神阙散中、高剂量组差异较为显著。对痛风性关节炎,神阙散高剂量组在用药后2 h即开始对痛风性关节炎产生药理作用,对模型大鼠关节炎肿胀指数具有显著降低作用,在用药后4~8 h治疗效果更加显著,其治疗效果与痛风定和秋水仙碱相当。病理结果显示,神阙散低、中、高剂量组均能改善模型大鼠痛风性关节炎的病理结果,且低、中、高剂量组呈现明显的量效关系。中剂量组与痛风定和秋水仙碱疗效相当,高剂量组改善作用优于痛风定和秋水仙碱。[结论]神阙散治疗痛风性关节炎疗效确切。

[Objective] To investigate the treatment effect of Shenque powder on rat gouty arthritis. [Methods] The rat model of hyperurieemia and gouty arthritis were built. By using full automatic biochemical analysis to observe rat＇ s disease activity index like Uric Acid （UA）, creatinine, urea nitrogen and swelling index to evaluate the pharmaeodynamics of Shenque powder on ratsgouty arthritis. [Results] The model is successful when the model group and control group had remarkable difference in both two groups. After taking medicine, in hyperurieemia model, the Uric Acid （UA）, ereatinine and urea nitrogen have statistical significance between the administration group and model group, the difference is significant with that in the high and median dose group. In gouty arthritis model, Shenque powder high dose group had pharmacological effects on gouty arthritis began after two hours in drug use, and arthritis rats swelling index was significantly reduced, especially between 4-8 h in drug use （P〈0.05）, the same treatment effect with Tongfengding and eolchieine. The Uric Acid （UA）, ereatinine, urea nitrogen, and dwelling index were still high in model group, other treatment group＇s disease index had decreased. As a result, the high dose group can improve pathology index of gouty arthritis, and the dose-effect relationship was shown among low, median and high dose. The median dose had the equal efficacy and the high dose had superior improving effect compared with Tongfengding and eolehicine. The Shenque powder is effective for gouty arthritis.
