摘要
目的 探讨Abhd2对慢性阻塞性肺疾病(chronic obstructive pulmonary disease, COPD)大鼠的肺功能和气道炎症的影响。方法 将20只Wistar大鼠随机分为正常对照组和COPD组,每组10只。采用熏吸香烟加气管内注入脂多糖的方法构建COPD模型。分别检测两组大鼠的肺功能FEV0.1、Abhd2 mRNA的表达、IL-6和IL-8及气道炎症细胞的变化,并分析Abhd2与肺功能FEV0.1及炎症因子IL-6和IL-8之间的相关性。结果 COPD大鼠与正常对照组大鼠相比,(1)肺功能指标FEV0.1[(0.68±0.02)ml,(0.73±0.04)ml]明显降低(t=3.386, P〈0.05);(2)肺组织Abhd2 mRNA的表达水平(0.26±0.07 vs 1.0)明显下降(t=5.831, P〈0.05)。(3)气道内炎症细胞明显浸润,以中性粒细胞、巨噬细胞和淋巴细胞为主。肺泡灌洗液中炎症因子IL-6[(403.68±42.67)ng/ml, (16.96±2.50)ng/ml]和IL-8[(1208.80±145.61)ng/ml, (43.49±4.92)ng/ml]明显增高(t=27.87, 24.69),P均〈0.05;肺组织中二者mRNA的表达水平(5.46±0.27, 16.35±1.24 vs 1.0)增高( t=8.256, 5.470),P均〈0.05,差异有统计学意义。(4)COPD大鼠肺组织中Abhd2 mRNA表达水平与肺功能FEV0.1、炎症因子IL-6和IL-8的表达水平均呈负相关(r1=0.892, P1=0.000;r2=0.697,P1=0.001; r3=0.682, P3=0.001)。结论 COPD大鼠Abhd2基因表达下降,上调Abhd2基因的表达可能成为改善肺功能、减轻气道炎症及治疗COPD的新途径。
Objective To investigate the correlation of Abhd2 with lung function and airway inflammation in rat model of Chronic Obstructive Pulmonary Disease. Methods Test is divided into two groups, including COPD group and control group. Lung function, the levels of Abhd2 mRNA, IL-6 and IL-8 were detected in two groups; Analysis the correlation of Abhd2 with FEV0.1, IL-6 and IL-8, respectively. Results(1)The values of FEV0.1 in COPD rats [(0.68±0.02)ml]were significantly lower than that in the control group [(0.73±0.04)ml] (t=3.386, P〈0.05). (2) Abhd2 mRNA expression in lung tissue in COPD rats were significantly decreased than that in control rats(t=5.831, P〈0.05). (3) Infiltration of inflammatory cells including neutrophils, macrophages and lymphocytes were observed in COPD rats. Expression of inflammatory cytokines (IL-6, IL-8) in the BALF were markedly higher in COPD group [(403.68±42.67)ng/ml, (1208.80±145.61)ng/ml, respectively] than in the normal group [(16.96±2.50)ng/ml,(43.49±4.92)ng/ml, respectively] (t=27.87, 24.69,P〈0.05). Similarly, in the lung tissue, two inflammatory cytokines were significantly increased(5.46±0.27, 16.35±1.24 vs 1.0)in the former than in the latter( t=8.256, 5.470, P〈0.05). (4) Abhd2 was negatively correlated with FEV0.1 as well as IL-6 and IL-8 in COPD rats (r1=0.892, P1=0.000;r2=0.697,P1=0.001; r3=0.682, P3=0.001). Conclusion Abhd2 expression was decreased in lung tissue in COPD rats. Up-regulation of Abhd2 might be a novel approach for improving lung function, reducing airway inflammation and treatment of COPD.
