摘要
目的探讨槲皮素对脑胶质瘤大鼠Bcl-2和Bax表达的影响。方法雌性Wistar大鼠30只,随机分为3组,即假手术组、模型组和槲皮素治疗组,每组10只。建立大鼠C6胶质瘤模型,于接种后17d处死各组大鼠,采用免疫组织化学法和Western blot法检测肿瘤组织Bcl-2和Bax蛋白表达水平的变化。结果与假手术组相比,模型组大鼠脑组织Bcl-2蛋白表达的平均光密度值显著升高,而Bax蛋白表达的平均光密度值显著降低(p<0.01);与模型组相比,槲皮素治疗组大鼠脑组织Bcl-2的蛋白表达的平均光密度值显著降低,而Bax蛋白表达的平均光密度值显著升高(P<0.01)。结论槲皮素能够降低脑胶质瘤大鼠Bcl-2的表达,上调Bax的表达。
Objective To study the effect of quercetin on the expressions of Bcl-2 and Bax in rats with brain glioma.Methods A total of 30 female rats were randomly divided into 3 group（n =10 for each）：sham group,model group and quercetin treatment group.The rat model of C6 glioma was established,and animals were killed 17 days after tumor graft.The expressions of Bcl-2 and Bax were observed by immunohistochemistry and Western blot.Results Compared with the sham group,the mean optic density（MOD） value of Bcl-2 positive expressions in brain tissues of the rats in model group was higher,but lower for Bax（P〈0.01）;and compared with the model group,the MOD value of Bcl-2 positive expressions in brain tissues of the rats in quercetin treatment group was lower,but higher for Bax（P〈0.01）.Conclusion Quercetin could down-regulate the expression of Bcl-2 and up-regulate the expression of Bax in rats with brain glioma.
