摘要
目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞中微小RNA-148a及靶基因DNA甲基转移酶1的表达变化及其临床意义。方法采集来自我院的31例SLE患者和22名健康对照者的外周抗凝血,定量PCR检测外周血单个核细胞中微小RNA-148a(hsa-miR-148a)及DNA甲基转移酶1mRNA水平,进一步分析hsamiR-148a的含量与DNA甲基转移酶1的表达和SLE患者疾病活动指数的相关性。结果 SLE患者的外周血中hsa-miR-148a的表达水平明显高于对照组(P<0.05),DNA甲基转移酶1的mRNA水平显著低于对照组(P<0.05),两者的含量呈显著负相关(P<0.001)。而且SLE患者的外周血细胞hsa-miR-148a表达与SLEDAI积分正相关(P<0.01)。结论 hsamiR-148a可能通过调节靶基因DNA甲基转移酶1的表达、降低细胞基因组甲基化水平从而参与SLE的发病,hsa-miR-148a可能成为一个新的SLE分子标志物及药物干预靶点。
Objective To investigate the expression and clinical significant of miR-148 a and its targeted gene DNA methyltransferase-1 in peripheral blood mononuclear cells of the patients with systemi clupus erythematosus(SLE).Methods The blood samples with anticoagulant were collected from 31 SLE patients and 22 normal persons.The expression of miR-148 a mRNA and its targeted gene DNA methyltransferase-1 mRNA in peripheral blood mononuclear cells was detected by real-time quantitative reverse transcriptase-polymerase chain reaction(qRT-PCR),and further to analyze the relationship between their expression and SLE activity index.Results Compared with the normal persons,the relative expression of hsa-miR-148 a in peripheral blood was increased in SLE patients(P〈0.05),the relative expression of DNA methyltransferase-1mRNA in peripheral blood was significantly decreased(P〈0.05).In addition,the expression of miR-148 a in peripheral blood of the patients was positively related to SLEDAI(P〈0.01).Conclusion HOTAIR Hsa-miR-148 a plays a critical role in decreasing DNA methylation in the development of SLE by targeting DNA methyltransferase-1,and miR-148 a might be a molecular marker and be used as a drug target.
