摘要
目的探讨miR-125b在胶质瘤中的表达及其与预后的相关性,分析其作为胶质瘤预后判断指标的可能性。方法采用荧光实时定量PCR方法检测miR-125b在不同级别胶质瘤及正常组织中的表达情况。建立受试者工作特征(ROC)曲线,计算ROC曲线下的面积(AUC)来评估miR-125b在胶质瘤组织和瘤旁正常组织中表达水平差异性。通过Kaplan-Meier法为高表达群体及低表达群体建立生存曲线,并用Log-rank检验评估两组生存曲线的差异。结果 miR-125b在胶质瘤组织中的表达水平显著高于正常组织,与胶质瘤的病理级别呈正相关。在生存分析中,miR-125b高表达水平的胶质瘤患者预后较差。结论 miR-125b是胶质瘤恶性程度的重要标志物,可做为胶质瘤级别分类及预后判断的生物学标记物。
Objective To study the expression and clinical significance of miR-125 b in human glioma tumor tissue and adjacent non-tumor tissue.Methods The expressions of miR-125 b were detected by qRT-PCR,Receiveroperating characteristic(ROC) curves were constructed,and the area under the ROC curves(AUC) was calculated to evaluate the significance of miR-125 b in distinguishing glioma tumor tissues from adjacent normal tissues.The survival curves of Kaplan-Meier method were constructed for both high expression group and low expression group,and the difference between curves was evaluated by log-rank test.Results The expression level of miR-125 b was higher in glioma tumor than in adjacent non-tumor tissues,positively correlated to glioma malignancy,with worse prognosis for the patient with high malignant glioma.Conclusion miR-125 b is closely correlated to the development and prognosis of human glioma.
