摘要
目的评价紫草素(shikonin)对斑马鱼的胚胎发育毒性及其血管抑制作用。方法将受精后0、10、24、48 h(hours post fertilization,hpf)发育正常的斑马鱼胚胎分别置于含有不同剂量紫草素的胚胎培养液中12、24、36、48 h后,显微镜下观察紫草素对斑马鱼胚胎的畸形率、死亡率、心率和血管生成的影响。结果给药24 h后,紫草素对不同发育阶段斑马鱼的LD50在0.73~1.56 mg/L之间。给药48 h后,该LD50值在0.25~0.39 mg/L之间。在给药大于0.5 mg/L时,紫草素能在12 h内引起斑马鱼的发育畸形,分别在1 h和24 h内均能引起斑马鱼心率衰减(P〈0.01),在12 h内可以显著抑制斑马鱼正常血管的生长。结论紫草素对斑马鱼胚胎发育具有毒性,并对斑马鱼血管生成具有一定的抑制作用。
AIM To evaluate shikonin on the developmental toxicity and vascular inhibition of zebrafish embryos. METHODS Post fertilization embryos of zebrafish were exposed to shikonin at a series concentrations for0,10,24,or 48 hours. The influence of shikonin on the embryos development and angiogenesis of zebrafish was observed with microscope; their embryo deformity rate,mortality rate and heart rate were also analyzed. RESULTS After treating for 24 h,the LD50 of shikonin on the embryo at the developmental stages ranged from 0. 73-1. 56 mg / L. After treating for 48 h,the LD50 of shikonin ranged from 0. 25-0. 39 mg / L. When treated at concentrations greater than 0. 5 mg / L,shikonin caused the embryo deformity in 12 h; decreased its heart beat in 1 and 24 h( P〈0. 01); inhibited the vessel growth of zebrafish in 12 h. CONCLUSION Shikonin has the toxic effects on the development of zebrafish embryos and the growth inhibitory effects on zebrafish's angiogenesis.
