植物抑菌剂抑菌机理的研究方法进展 被引量:9

摘要 综述了植物抑菌剂作用机理研究中常用的几种技术手段,从植物抑菌剂对微生物细胞形态的影响、对细胞壁、细胞膜、能量代谢和遗传物质的作用等不同方面加以介绍。采用SEM、TEM、AFM和CLSM方法观察微生物的形态结构;从细胞膜的通透性、流动性、结构变化和内溶物外流4个方面检测细胞膜的变化;采用呼吸代谢抑制试验、Clark氧电极法和荧光猝灭试验方法研究植物抑菌物质对菌体呼吸代谢的影响;论述光谱分析法、电化学方法、核磁共振法、计算机模拟技术手段对抑菌剂与DNA的相互作用方式的检测与分析。并对植物抑菌剂抑菌机理的分析方法的发展应用进行了展望。 Some common antibacterial mechanism methods of plant bacteriostat were discussed in this paper, which introduced the influences on microbial cell morphology, cell membrane, cell wall, cellular respiration metabolism as well as the genetic material. The methods of SEM, TEM, AFM and CLSM observing microbial morphology were applied; the detection methods were illustrated in the four aspects of cell membrane structure, permeability, liquidity and the flow of solute^the respiratory metabolism inhibition test,Clark oxygen electrode method and fluorescence quenching test method were used to study the influence of plant bacteriostat on respiratory metabolism; spectral analysis method,electrochemical method, nuclear magnetic resonance (NMR) method, computer simulation technology were used to detected the interactions between antibacterial agent and DNA. This article mainly reviewed the current methods and means on various microbial structures, and evaluated briefly. Problems and prospects of the research and development of antibacterial mechanism methods were also presented.
