丁香降气方对反流性食管炎大鼠IL-23/IL-17炎症轴的影响 被引量:15

Influence of the “Dingxiang Jiangqi Decoction” on IL-23 / IL-17 inflammation axis in rats with reflux esophagitis

摘要 目的观察丁香降气方对反流性食管炎大鼠IL-23/IL-17炎症轴水平的影响。方法将40只SD大鼠随机分为正常组、模型组、中药组及西药组,每组10只。除正常组外均行贲门肌切开术加幽门半结扎术制备反流性食管炎模型,并分别给予相应干预措施。光镜下观察各组大鼠食管组织形态学变化,ELISA法及RT-PCR法检测IL-23 mRNA、IL-17 mRNA、IL-17蛋白的表达。结果与正常组比较,各组食管黏膜均有不同程度炎症表现,但中药组较西药组轻微(P<0.05)。与正常组比较,模型组IL-17蛋白、IL-23mRNA及IL-17 mRNA含量明显升高(P<0.05)。与模型组比较,中药组IL-17蛋白、IL-17 mRNA、IL-23 mRNA降低(P<0.05),西药组IL-23 mRNA降低(P<0.05)。与西药组比较,中药组IL-17蛋白含量降低、IL-17 mRNA表达下降(P<0.05)。中药组IL-17蛋白含量与正常组比较差异无统计学意义(P>0.05)。中药组、西药组IL-23 mRNA表达与正常组比较差异无统计学意义(P>0.05);同时中药组与西药组IL-23 mRNA表达比较差异无统计学意义(P>0.05)。结论丁香降气方能有效改善反流性食管炎大鼠的食管黏膜炎症,其对IL-23/IL-17炎症轴的调控可能是其作用机制之一。 Objective To observe influence of the"Dingxiang Jiangqi Decoction"on IL-23 / IL-17 inflammation axis in rats with reflux esophagitis. Methods 40Spregue-Dawley( SD) rats were randomly divided into treatment group,control group,model group and normal group,10 in each group. Reflux esophagitis models were induced by cardiomyotomy and pylorus semiligation,and given the corresponding intervention measures. Pathological changes of esophagus tissues were observed by light microscope in each group,and the expression of IL-23,I L-17 mRNA in esophageal tissues of rats were detected by real-time RT-PCR,IL-17 protein content in esophageal tissues was measured by ELISA. Results Compared with the normal group,the esophageal mucosa in each group had different degrees of inflammation,but the Dingxiang Jiangqi Decoction group was mild than the western medicine group( P 0. 05). The expressions of IL-23 and IL-17 mRNA and IL-17 protein in the esoph ageal tissues of model group were significantly higher than the normal group( P 0. 05). Compared with model group,the expressions of IL-23 and IL-17 mRNA and IL-17 protein in the esoph ageal tissues of Chinese medicine group were lower( P 0. 05),and the expression of IL-23 mRNA in western medicine was lower( P 0. 05). Compared with western medicine group,IL-17 protein decreased and IL-17 mRNA expression decreased( P 0. 05). There was no significant difference of IL-17 protein expression between the Dingxiang Jiangqi Decoction group and the normal group( P 0. 05).There was no significant difference in IL-23 mRNA expression in Dingxiang Jiangqi Decoction group,control group and normal group( P 0. 05); while there was no significant difference between the Dingxiang Jiangqi Decoction group and the western medicine group( P 0. 05). Conclusion "Dingxiang Jiangqi Decoction"can effectively improve the esophageal mucosal inflammation of reflux esophagitis rats,the regulation of the IL-23 / IL-17 inflammation axis may be one of the mechanisms of the"Dingxiang Jiangqi Decoction".
