摘要
目的:观察番茄红素对大鼠心肌梗死后心功能及心肌细胞凋亡的影响。方法:通过结扎大鼠冠状动脉前降支制备心肌梗死模型,分别给予番茄红素及生理盐水灌胃。28d后行心脏超声及血流动力学检测心功能,Tunel染色检测心肌细胞凋亡,Western blot法检测梗死周边Caspase-3蛋白表达。结果:心梗后,LVEF、FS、LVESP、-dP/dt、+dP/dt显著降低(P<0.05),LVEDD、LVESD、LVEDP显著升高(P<0.05),心肌细胞凋亡指数显著上升(P<0.05),梗死周边区Caspase-3蛋白表达水平显著上调。与心梗对照组相比,番茄红素治疗组LVEF、FS、LVESP、-dP/dt、+dP/dt显著升高(P<0.05),LVEDD、LVESD、LVEDP显著降低(P<0.05),心肌细胞凋亡指数显著下降(P<0.05),梗死周边区Caspase-3蛋白表达水平下调。结论:番茄红素可能通过抑制心梗后心肌细胞凋亡逆转心室重构,从而改善心功能。
Objective: To observe the effect of lycopene on cardiac function and myocardial apoptosis af- ter rat's myocardial infarction(MI). Methods: Sprague-Dawley rat MI model was established by left anterior descending coronary artery ligation. After operation, rats received lycopene or sa- line. After 28 days, cardiac function was assessed by cardiac ultrasound and hemodynamics tech- nology. Myocardial apoptosis was detected by Tunel staining. Caspase-3 protein expression was detected in peri-infarcted zone by Western blot. Results: After MI, LVEF, FS, LVESP and dP/dt decreased significantly (P〈0.05), while LVEDD, LVESD, LVEDP and myocardial apop- tosis index increased significantly (P〈0.05). Caspase-3 protein expression was significantly up- regulated in peri-infarcted zone. Lycopene improved cardiac function, inhibited the apoptosis in- dex, and up-regulated Caspase-3 protein expression. Conclusion: Lycopene may inhibit myocardi- al apoptosis and ventricular remodeling, which improves cardiac function.
