摘要
目的:研究ETNPPL是否能够影响肝细胞癌细胞系的增殖表型如细胞活性和克隆形成。方法:Western blot检测ETNPPL在不同肝细胞癌细胞系中的基础表达量后,设计并构建ETNPPL的靶向siRNA,验证敲低效果,采用CCK-8实验和平板克隆形成实验,观察细胞的增殖能力。结果:与对照组相比,干扰ETNPPL的表达可以显著抑制肝细胞癌细胞系97H和LM3的增殖能力(P<0.05)和克隆形成能力(P<0.05)。结论:ETNPPL具有调控肝细胞癌细胞增殖和克隆形成能力的功能;ETNPPL可能是治疗肝细胞癌的一个潜在靶点。
Objective: To investigate the role of ETNPPL in proliferation of hepatocellular carcinoma. Methods: The expression level of ETNPPL in different hepatocellular carcinoma cell lines was mesearsed by Western blot. siRNA targeting ETNPPL was designed, synthetized and transfected into the cells. After the validation of efficiency of knock-down, the proliferation rates and ability of colony forming were measured by CCK-8 assay and plate colony forming assay respectively. Results: Compared with the control group, after the knock-down of ETNPPL, the proliferation rates and the ability of colony forming of 97H and LM3 cells both significantly reduced (P〈 0.05). Conclusion: ETNPPL can modulate the proliferation and the ability of colony forming and may function as a potential target in hepatocellular carcinoma treatment.
