子宫内膜腺癌雌激素受体亚型的表达及其与临床病理的关系 被引量:1

Expression of estrogen receptor subtype and its relationship with clinicopathology in adenocarcinoma of endometrium

摘要 目的探讨雌激素受体α(ERα)、ERβ在子宫内膜样腺癌组织中的表达及其与临床病理特征的关系。方法选择2011年1月~2012年12月在广西医科大学附属肿瘤医院经组织学证实的93例子宫内膜腺癌(子宫内膜癌组)及31例正常子宫内膜(对照组)组织,分别采用qRT-PCR及免疫组化法检测ERα、ERβ mRNA及蛋白的表达情况。结果子宫内膜癌组ERα mRNA表达量及蛋白阳性表达率、ERα/ERβ mRNA均明显低于对照组,差异均有统计学意义(P〈0.05或P〈0.01);随着临床分期、组织学分级、肌层浸润加深,ERα mRNA表达量、蛋白阳性表达率及ERα/ERβ mRNA比值逐渐下降(P〈0.05或P〈0.01);子宫内膜癌组ERβ mRNA及蛋白表达也降低,与对照组比较,差异均无统计学意义(均P〉0.05)。子宫内膜癌组织中ERα与ERβ阳性表达率呈负相关(r=-0.234,P〈0.05)。结论在子宫内膜癌的发生发展中,ERα的变化可能较ERβ更有意义,ERα/ERβ mRNA比值可作为判断子宫内膜癌预后的指标。 Objective To explore the expression of estrogen receptor alpha(ERα) and beta(ERβ) and its relationship with clinical and pathological characteristics in tissues with adenocarcinoma of endometrium. Methods 93 cases of endometrial adenocarcinoma(endometrial adenocarcinoma group) and 31 cases of normal endometrium(control group)from January 2011 to December 2012 in Affiliated Tumor Hospital of Guangxi Medical University were selected, Realtime fluorescence quantitative PCR(qRT-PCR) and immunohistochemistry were used to detect the expression of ERαand ERβ in endometrial tissues of two groups. Results Expression of ERα mRNA and protein, ERα/ERβ mRNA were significantly lower in endometrial adenocarcinoma group than that in control group(P〈0.05 or P〈0.01); with the increasing of clinical stages, histological grades and depth of myometrial invasion, the expression of ERα mRNA and protein, ERα/ERβ mRNA were significantly reduced(P〈0.05 or P〈0.01); compared with the control group, expression of ERβ mRNA and protein in endometrial adenocarcinoma group were reduced, the difference was no statistically significant(P〉0.05). There was a negative correction between ERα and ERβ in endometrial adenocarcinoma(r =-0.234,P〈0.05). Conclusion ERα may play a different and more important role in the occurrence and development of endometrial carcinoma compared to ERβ, ERα/ERβ mRNA ratio can be used to judge prognosis of endometrial carcinoma.
