芝麻素对高糖条件下PC12细胞Aβ和BACE-1表达的影响 被引量:1

Effects of Sesamin on Aβ and BACE-1 Expression in PC12 Cells in High Glucose Culture Conditions

摘要 目的探讨芝麻素对高糖条件下鼠嗜铬细胞瘤细胞(PC12细胞)β位淀粉样前体蛋白裂解酶-1(BACE-1)和β淀粉样蛋白(Aβ)代谢的影响。方法培养PC12细胞,随机分为5组:对照组(正常培养组)、高糖培养组(25mmol/L)、高糖(25 mmol/L)加芝麻素0.5μmol/L组、高糖(25 mmol/L)加芝麻素5.0μmol/L组和高糖(25 mmol/L)加芝麻素50μmol/L组,酶联免疫吸附试验(ELISA)检测Aβ1-42水平,荧光实时定量PCR(RT-PCR)检测BACE-1 mRNA表达,Western blot检测BACE-1蛋白表达水平。结果高糖培养的PC12细胞组的Aβ1-42水平明显高于正常条件培养的PC12细胞组(P<0.05);在加入0.5、5.0和50μmol/L芝麻素后,Aβ水平显著降低(P<0.05);高糖培养的PC12细胞组BACE-1 mRNA和蛋白表达显著高于正常培养PC12细胞组(P<0.05);在加入0.5、5.0和50μmol/L芝麻素后,BACE-1 mRNA和蛋白水平显著降低(P<0.05)。结论在高糖条件下,PC12细胞Aβ1-42、BACE-1 mRNA和蛋白表达明显升高,芝麻素浓度依赖性地降低高糖环境下PC12细胞中升高的Aβ1-42水平,降低BACE-1 mRNA和蛋白表达。 Objective To explore the effects of Sesamin on the BACE-1 and Aβ expression in Rat Pheochromocytoma cells (PC12 ceils) in high glucose culture conditions. Methods The PC12 cells were randomly divided into five groups: Control group (PC12 cells in normal culture conditions), PC12 cells in high glucose culture conditions (25 mmol/L), PC12 cells in high glucose conditions (25 mmol/L) plus Sesamin 0. 5 μmol/L,5.0 μmol/L and 50 μmol/L. Aβ1-42 concentration was measured by ELISA. BACE-1 mRNA and protein expression was measured by RT-PCR and Western blot. Results Compared with PC12 cells under normal conditions, the Aβ1-42 level, the BACE-1 mRNA and protein expression were elevated in PCI2 cells in high glucose conditions (P 〈 0. 05 ) ;meanwhile, the AIM-42 concentration, BACE-1 mRNA and protein ex- pression in high glucose was significantly decreased after intervened by 0. 5,5.0 and 50 I.Lmol/L Sesamin ( P 〈 0. 05 ). Conclusion The Aβ1-42, BACE-1 mRNA and protein expression are increased in PC12 cells in high glucose conditions, Sesamin concentration-dependently reverse the increasing Aβ1-42 ,BACE-1 mRNA and protein level in PC12 cells in high glucose conditions.
