摘要
目的:探讨氧化苦参碱对多发性骨髓瘤(MM)U266细胞凋亡的诱导作用及机制。方法:以不同终浓度(0、5、10、20mmol/L)氧化苦参碱处理MM U266细胞。分别采用MTT法、瑞特染色、流式细胞仪及western blot检测MM U266细胞增殖、细胞凋亡形态、细胞凋亡及其凋亡因子(Bax、Bcl-2)蛋白表达。结果:氧化苦参碱(5、10、20mmol/L)可显著抑制MM U266细胞增殖,呈剂量依赖性。20 mmol/L氧化苦参碱作用48h,MM U266细胞呈现典型凋亡形态学改变。氧化苦参碱(5、10、20mmol/L)作用48h后,MM U266细胞凋亡率分别为(11.5±1.4)%、(24.7±2.5)%、(43.6±3.4)%,呈剂量依赖性。氧化苦参碱(5、10、20mmol/L)作用MM U266细胞48h,可上调Bax蛋白表达,下调Bcl-2蛋白表达,并呈现剂量依赖性。结论:氧化苦参碱可诱导MM U266细胞凋亡,调控凋亡因子Bax、Bcl-2的表达是其可能作用机制。
Objective:To evaluate the apoptosis of multiple myeloma(MM)cell line U266 induced by oxymatrine and its mechanism.Method:MM cell line U266 were treated with different concentrations(0,5,10or20mmol/L)of oxymatrine.The cell proliferation,cell apoptotic morphology,cell apoptosis and protein expression of apoptosis factors(Bax and Bcl-2)were respectively determined by MTT method,Wright staining,flow cytometry and western blot.Result:Oxymatrine(5,10 or 20 mmol/L)significantly inhibited the cell proliferation of U266,and showed a dose dependent effect.After treatment of 20mmol/L oxymatrine for 48 h,U266showed typical apoptotic morphological changes.After treated with oxymatrine(5,10 or 20mmol/L)for 48 h,the apoptosis rate of U266 was respectively(11.5±1.4)%,(24.7±2.5)% and(43.6±3.4)%,and showed a dose dependent effect.After U266 were treated with oxymatrine(5,10 or 20 mmol/L)for 48 h,the expression of Bax increased and Bcl-2decreased,both dose dependently.Conclusion:Oxymatrine can induce the apoptosis of MM cell line U266.Regulating the expression of apoptosis related gene Bax and Bcl-2may be its mechanism.
