三七总皂苷联合雷公藤多苷对关节炎大鼠外周血TNF-α、IL-6、MCP-1以及胸主动脉MCP-1表达的影响 被引量:13

摘要 目的观察中药有效成分三七总皂苷联合雷公藤多苷对胶原诱导性关节炎(CIA)大鼠模型外周血肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、单核细胞趋化蛋白-1(MCP-1)以及胸主动脉MCP-1表达的影响。方法从50只大鼠中随机选取8只作为正常组,其余大鼠建立CIA模型。将成模大鼠随机分为模型组、雷公藤多苷组、联合组、甲氨蝶呤组,每组8只。雷公藤多苷组给予雷公藤多苷10 mg/(kg·d)灌胃,联合组给予三七总皂苷200 mg/(kg·d)和雷公藤多苷10 mg/(kg·d)灌胃,甲氨蝶呤组给予甲氨蝶呤1.6mg/(kg·d)灌胃,正常组和模型组给予等量生理盐水灌胃,均1次/d,共42 d。灌胃结束后次日应用液相蛋白芯片技术检测血清中TNF-α、IL-6、MCP-1水平,应用免疫组织化学法检测胸主动脉MCP-1的表达量。结果模型组血清中TNF-α、IL-6、MCP-1水平均明显高于正常组(P均<0.05),各给药组均明显低于模型组(P均<0.05),联合组均明显低于甲氨蝶呤组和雷公藤多苷组(P均<0.05),甲氨蝶呤组和雷公藤多苷组比较差异无统计学意义(P均>0.05)。模型组MCP-1表达量明显低于正常组(P<0.05),各给药组MCP-1表达量均明显高于模型组(P均<0.05),联合组明显高于甲氨蝶呤组和雷公藤多苷组(P均<0.05),甲氨蝶呤组和雷公藤多苷组比较差异无统计学意义(P>0.05)。结论三七总皂苷联合雷公藤多苷可明显降低血清炎症因子TNF-α、IL-6、MCP-1水平,上调MCP-1在胸主动脉的表达,且较单用雷公藤多苷或甲氨蝶呤效果更优,提示中药有效成分配伍在抗炎、保护心血管方面具有增效作用。 Objective It is to detect the active ingredients of traditional Chinese medicine PNS combined with Tripterygium glycosides( TG) on tumor necrosis factor( TNF- α),interleukin- 6( IL- 6),monocyte chemotactic protein- 1( MCP- 1)levels in serum of CIA rats. Methods 50 rats were selected in which 8 rats as normal group and the other rats were used to establish CIA models,then the models were randomly divided into model group,TG group,combined group,Methotrexate group( MTX),each group included 8 samples. Normal group and model group were given equivalent volume of saline. TG group was given TG 10 mg /( kg·d) by gavage,combined group given TG 10 mg /( kg·d) + PNS 200 mg /( kg·d) by gavage,MTX group given MTX 1. 6 mg( kg·d),all the groups were treated for 49 days. On 49 nd day after modeling,protein chip technology was used to measure TNF- α,IL- 6,MCP- 1 levels in serum,the expression of MCP- 1 in thoracic aorta were detected by immunohistochemical method. Results The levels of serum TNF- α,IL- 6,MCP- 1 in model group were higher than that in normal group( P all 0. 05),the levels in every treated group were lower than that in model group( P all 0. 05),those in combined group were lower than that in TG group and MTX group( P all 0. 05). No significant difference was found between TG group and MTX group( P〈0. 05). The expression of MCP- 1 in thoracic aorta was lower in model group than that in normal group( P all 0. 05),the levels in every treated group were higher than that in model group( P all 0. 05),those in combined group were higher than that in TG group and MTX group( P all 0. 05). No significant difference was found between TG group and MTX group( P〉0. 05). Conclusion PNS + TG can significantly reduce the levels of TNF-α,IL- 6,MCP- 1 in serum,up-regulate the level of MCP- 1 in thoracic aorta,and the effect was more significantly than simple drug,which indicate that compatibility of traditional Chinese medicine active ingredients has a synergistic effect on antiinflammatory and the aspect of prevention of cardiovascular damage.
