HPLC法同时测定甘草总黄酮分散片中甘草素和甘草查尔酮A的含量 被引量:9

Simultaneous Determination of Liquiritigenin and Licochalcone A in Licorice Flavonoids Dispersible Tablets by HPLC

摘要 目的建立同时测定甘草总黄酮分散片中甘草素和甘草查尔酮A含量的方法。方法采用高效液相色谱法,Inertsil C18柱(4.6 mm×150 mm,5μm),流动相以乙腈(A)-0.1%磷酸水(B)梯度洗脱(0-8 min,25%-32%A;8-30 min,32%-45%A;30-42 min,45%-60%A;42-50 min,60%-90%A),柱温30℃,检测波长为276 nm,365 nm,流速1.0 mL/min。结果甘草素和甘草查尔酮A的线性范围分别为0.016 1-0.644 0μg(r=0.999 8),0.019 3-0.771 2μg(r=0.9997)。甘草素平均回收率为99.16%,RSD=1.51%;甘草查尔酮A平均回收率为98.48%,RSD=1.22%。结论该法简便、灵敏度高、重现性好,可作为甘草总黄酮分散片中指标性成分的含量测定和成品质量控制。 OBJECTIVE To establish a method for simultaneous determination of liquiritigenin and licochalcone A in Licorice flavonoids dispersible tablets by HPLC. METHODS Inertsil C18column( 4. 6 mm × 150 mm,5 μm) was used with mobile phase of Acetonitrile-0.1% H3PO4 soiution gradient elution( 0-8 min,25%-32% A; 8-30 min,32%-45% A; 30-42 min,45%-60% A; 42-50 min,60%-90% A). The detection wavelength was set at 276 nm,365 nm. The flow rate was 1.0 mL / min and the column temperature was maintained at 30 ℃. RESULTS The linear relationship of liquiritigenin and licochalcone A were 0.016 1-0.644 0 μg( r = 0.999 8),0.019 3-0.771 2 μg( r = 0.999 7). The average recovery rate were 99. 16% with RSD 1. 51%; 98. 48% with RSD 1. 22%( n = 6),respectively.CONCLUSION The established method is simple,accurate and reliable. It can be used for determination of the 2components and quality control in Licorice flavonoids dispersible tablets.
