摘要
试验旨在探究不同浓度褪黑素刺激对内蒙古绒山毛囊干细胞增殖活性的影响。采用0(对照组)、100、300、500、700pg/mL 5个浓度梯度分别对内蒙古绒山羊毛囊干细胞连续刺激5d,MTT法分别检测各组细胞活性。结果表明,不同浓度褪黑素刺激2d后,相较于对照组(0pg/mL),褪黑素刺激组未见成团细胞出现,开始出现长梭形细胞且细胞生长不再均一,各刺激组均表现出较高的增殖活性;刺激4d后,500pg/mL褪黑素刺激组毛囊干细胞增殖活性显著高于0、100pg/mL(P<0.01;P<0.05),但与700pg/mL褪黑素刺激组差异不显著(P>0.05)。因此,500pg/mL是内蒙古绒山羊毛囊干细胞增殖的最适浓度。
This study aimed to investigate the effect of different concentrations of melatonin on proliferative activity of hair follicle stem cells in Inner Mongolia cashmere goat.The five levels melatonin including 0,100,300,500 and 700pg/mL were designed to stimulate the hair follicle stem cells in Inner Mongolia cashmere goat,the MTT method was used to detect cell proliferation activity for 5days.The result showed that 2days later,the groups stimulated by melatonin did not cobble-stone arrangement and began to appear spindle cell and cell growth was not uniform compared with the control group(0pg/mL),the groups showed obvious effect on cell proliferation relatively.4days later,the cell proliferation of groups stimulated by 500pg/mL melatonin were significantly greater than that of 0,100pg/mL(P〈0.01;P〈0.05),but there was no significant difference with 700pg/mL melatonin stimulation group(P〈0.05).The optimal melatonin concentration of hair follicle stem cell proliferation was 500pg/mL in Inner Mongolia cashmere goat.
