摘要
目的探讨RNA结合蛋白QKI-5在直肠癌组织中的表达及其与正常直肠组织的表达差异。方法收集新鲜手术切除标本通过免疫印迹和Real-time PCR方法观察直肠癌组织与正常直肠黏膜组织间QKI-5的表达差异,使用相同方法观察不同分化水平直肠癌组织中QKI-5的表达变化趋势。通过所收集标本的病例资料对RNA结合蛋白QKI-5与多种临床病理特征进行统计学分析。结果 Western blot实验结果显示,在直肠癌组织中QKI-5的表达水平显著低于正常直肠组织(P<0.05),于此同时Real-time PCR实验也得到了类似结果(P<0.05)。通过Western blot比对不同分化水平的直肠癌组织发现,随着分化程度的不断降低QKI-5的表达水平也随之降低(r=-0.917,P<0.05)。通过与临床病理特征间的比对QKI-5与肿瘤TNM分期、淋病结转移、远端转移、肿瘤标志物CEA密切相关(P<0.05)。结论 QKI-5在直肠癌发生发展中很可能起到了抑癌基因的作用,针对QKI-5为靶点的研究对于构建早期诊断直肠癌分子标志物和开拓直肠癌生物治疗靶点都有着重要意义。
Objective To investigate the differences of expression of RNA-binding protein of QKI-5 between eolorectal cancer tissues and normal rectal tissues. Methods The fresh surgical ablation specimens of colorectal carcinoma were collected, and the expression levels of QKI-5 were detected and compared by immunoblotting and Real-time PCR methods. The expression levels of QKI-5 in different differentiation levels of colorectal carcinoma tissues were observed by using the same method. The correlation between clinicopathological characteristics of colorectal cancer and QKI-5 was statistically analyzed. Results Western Blot results showed that the expression levels of QKI-5 in colorectal cancer tissues were significantly lower than those in normal rectal tissues ( P 〈 0.05 ), meanwhile, Real-time PCR also got similar results (P 〈0.05). With the continous decrease of differentiation levels of colorectal cancer tissue, the expression levels of QKI-5 were correspondingly decreased ( r = -0. 917, P 〈 0. 05 ). Furthermore QKI-5 was closely correlated to TNM staging, lymph node metastasis, distant metastasis and tumor marker - CEA ( P 〈 0.05 ). Conclusion QKI-5 may play an important role of anti - oncogene in the pathogenesis and development of colorectal carcinoma, thus, the researches taking QKI-5 as target point are of important significance for the construction of molecular biological markers in early diagnosis and research & development of biotherapeutic targets of colorectal carcinoma.
