桑白皮通过Rap1-MAPK信号途径抑制钙内流促进新生大鼠皮质神经元的分化研究 被引量:2

摘要 目的通过分析桑白皮提取物对新生大鼠神经元分化的影响,探究桑白皮提取物促进神经元分化的机制。方法制备桑白皮提取物作用于原代培养的新生鼠皮质神经元细胞,通过形态学观察神经元突起生长情况;利用蛋白免疫印迹实验检测与神经元分化信号通路相关的Rap1和ERK1/2的表达,通过钙影像实验分析桑白皮提取物对神经元细胞内钙离子浓度的影响。结果桑白皮提取物促进神经元突起的分化,并增加Rap1-GTP和磷酸化ERK1/2蛋白的表达,同时能有效抑制神经元钙离子内流。结论桑白皮提取物促进新生大鼠皮质神经元分化,其促进作用可能是通过调控Rap1-ERK1/2信号通路以及抑制细胞钙离子内流实现的。
