加味桂枝茯苓颗粒对大鼠良性前列腺增生症微血管密度计数及bcl-2基因表达的影响 被引量:2

Influence of modified Guizhi Fuling Granules on MVD counting and expression of bcl-2 gene in rats with benign prostatic hyperplasia

摘要 目的:了解加味桂枝茯苓颗粒对大鼠良性前列腺增生症微血管密度(MVD)计数、bcl-2基因表达的影响,以期明确加味桂枝茯苓颗粒对前列腺新生血管产生和基质细胞生长干预的分子生物学机制。方法:大鼠80只,分为空白对照组,空白增生组,加味桂枝茯苓颗粒组和非那雄胺组。造模成功后,对应组分别采用不给药,0.9%氯化钠溶液,加味桂枝茯苓颗粒悬液,非那雄胺溶液灌胃治疗。后观察前列腺组织生长情况,免疫组化检测前列腺组织MVD,FCM检测bcl-2基因表达。结果:加味桂枝茯苓颗粒组和非那雄胺组较空白增生组MVD计数、bcl-2表达量为均显著下降(P<0.01),且两组之间无明显差异。结论:加味桂枝茯苓颗粒可明显降低大鼠前列腺MVD计数、bcl-2的基因表达,减少前列腺组织微血管产生,加速腺细胞的凋亡,明显减少基质细胞产生,可能是其治疗前列腺增生的重要原因。 Objective:To study the influence of modified Guizhi Fuling Granules on microvessel density(MVD) counting and expression of bcl-2 gene in prostate tissue of rats with benign prostatic hyperplasia,and clarify the molecular biological mechanism of modified Guizhi Fuling Granules in intervening angiogenesis and growth of stromal cells in prostate.Methods:80 rats were randomly divided into blank control group,blank hyperplasia group,modified Guizhi Fuling Granule group and finasteride group.After model establishing,rats in each group received corresponding treatments.The growth situation of prostate tissue of rats in each group was observed,the expression of MVD in prostate tissue was detected by using immunohistochemical technique,and the expression of Bcl-2 gene in prostate tissue was detected by using FCM.Results:The difference in the two indicators among modified Guizhi Fuling Granule group,finasteride group and blank control group was significant(P〈0.01),while there was no significant difference between modified Guizhi Fuling Granule group and finasteride group.Conclusion:Modified Guizhi Fuling Granule could significantly decrease MVD counting,expression of bcl-2 gene and angiogenesis,accelerate the apoptosis of gland cells,and reduce the producing of stromal cells in prostate tissue of rat,which may be an important mechanism for modified Guizhi Fulinq Granule in treating benign prostatic hyperplasia.
