摘要
目的:对比分析环丙沙星(CIP)、乳铁蛋白多肽嵌合体(LFchimera)单独使用和二者联合使用处理铜绿假单胞菌对其生物膜及密度感知(QS)系统信号分子(AHL)生成的抑制作用。方法:以铜绿假单胞菌野生菌株PAO1作为实验菌株,随机分为5组,分别为对照组(未做任何处理),实验组分为CIP(0.04μg/ml)处理组、LFchimera(0.25μmol/L)处理组、LFchimera(1.00μmol/L)处理组、CIP(0.04μg/ml)+LFchimera(1.00μmol/L)处理组,分别定量测定且比较各组PAO1生物被膜和QS系统信号分子表达情况。结果:CIP组、LFchimera(0.25μmol/L)组、LFchimera(1.00μmol/L)组、CIP+LFchimera组与对照组相比;LFchimera(1.00μmol/L)组与CIP组/LFchimera(0.25μmol/L)组相比;CIP+LFchimera组与其他所有组相比,POA1生物膜和QS系统信号分子表达均下降,差异有统计学意义(P<0.05)。CIP组与LFchimera(0.25μmol/L)组相比,差异无统计学意义(P>0.05)。结论:CIP和LFchimera联用抑菌作用增强,此抑制作用可能与抑制QS系统信号分子有关。
Objective:To compare and analyze the Ciprofloxacin(CIP) and Lactoferrin Polypeptide Chimera(LFchimera) used alone and combined use of pseudomonas aeruginosa inhibition of biofilm and quorum sensing(QS) system signaling molecules(AHL) were generated.Method:Pseudomonas aeruginosa strain PAO1 was used as the control group(untreated),CIP(0.04 μg/ml) group,LFchimera(0.25 μmol/L) group,LFchimera(1.00 μmol/L) group,CIP(0.04μg/ml)+LFchimera(1.00 μmol/L) group,the expression of POA1 and QS system were checked.Result:CIP group,LFchimera(0.25 μmol/L) group,LFchimera(1.00 μmol/L),CIP+LFchimera group and control group,LFchimera(1.00 μmol/L) group and CTP group/LFchimera(0.25 μmol/L) group,CIP+LFchimera group and other groups,the expression of POA1 and QS system were significantly decreased,the differences were significant(P〈0.05).There was no significant difference between CIP group and LFchimera(0.25 μmol/L) group,the difference was no statistically significant(P〉0.05).Conclusion:The inhibitory effect of CIP and LFchimera was enhanced,which may be related to the inhibition of QS signaling molecules.
