人胆囊癌裸鼠移植瘤模型的建立及超声观察

摘要 目的建立人胆囊癌裸鼠皮下移植瘤模型,并对其进行超声观察。方法培养GBC-SD细胞,将细胞浓度调至1×107/ml,取0.2 ml注射于裸鼠皮下。成瘤后作为瘤源,瘤块剪成约2 mm3的组织块,置入裸鼠皮下,待其成瘤。以后各代模型的瘤源均来自上一代模型,用同样的方法接种。记录瘤块成瘤率、成瘤时间、生长特点。观察瘤块外观,并进行超声、解剖及病理观察。结果细胞注射法及组织块法可成功建立胆囊癌裸鼠移植瘤模型,成瘤率为100%;各代组织块法之间成瘤时间差异无统计学意义(P>0.05);二维超声测量瘤体最大径与大体游标卡尺测量差异有统计学意义(P<0.05)。结论将细胞注射法与组织块法结合,是一种理想的建立人胆囊癌裸鼠移植瘤模型的实验方法。 Objective To establish the human gallbladder cancer nude-mouse transplanted tumor model and to observe by ultrasound. Methods GBC-SD cells were cultured and the cell concentration was adjusted to 1×10^7/ml,of which 0.2ml was subcutaneously injected to nude mouse.The cultivated tumor was selected as the tumor source and cut into tissue blocks of 2 mm3,which were then implanted into the subcutaneous tissue of nude mouse to cultivate tumor.The tumors of later generation were all cultivated from sources of tumors of the former generation by the same method.The tumor formation rate,time of tumor formation,and features of tumor formation were recorded.The appearance of tumor block was observed and ultrasonic,anatomical and pathological observations were conducted. Results The method of cell injection and tissue block implantation could successfully establish the gallbladder cancer nude-mouse transplanted tumor model,with tumor formation rate of 100%.The tumor formation time of each generation showed no statistically significant difference(P〉0.05).The maximum diameter of tumor detected by ultrasound and vernier caliper showed statistically significant differences(P〈0.05). Conclusion Combination of cell injection and tissue block implantation is a favorable method to establish the gallbladder cancer nude-mouse transplanted tumor model.
