犬子宫内膜上皮细胞和基质细胞的分离培养与鉴定

Isolation,Culture and Identification of Canine Endometrial Epithelial Cells and Stromal Cells

为了探究犬子宫内膜细胞的培养方法,采用组织块培养法和酶消化培养法进行细胞培养并使用倒置显微镜观察细胞的形态结构和生长状态,对细胞进行免疫组化鉴定。结果表明,组织块培养法需要1周左右培养出犬子宫内膜上皮细胞,且细胞不易纯化;酶消化培养法节省时间,获得的细胞活力强、纯度高。综上所述酶消化培养法更适于培养犬子宫内膜细胞,为后期细胞水平研究奠定基础。

To study the methods for culturing canine endometrial cells, tissue block culture method and enzymatic digestion culture method were used in this study. Cells were identified by immunohistochemical staining, the morphological structure and growth status of endometrial epithelium cells were observed by inverted phase-contrast microscope. Our results showed that tissue block culture method which is not easy to purify cells took longer time to get endometrial epithelium cells than the enzymatic diges- tion culture method .The enzymatic digestion culture method got high-viability and high-purity cells in 24 hours. In conclusion, en- zymatic digestion culture method is appropriate for canine endometrial cell culture, which can be used for the further study.
