花生ABI3基因的克隆、序列分析及表达载体构建 被引量:2

摘要 脱落酸(ABA)作为植物体内的一种重要激素,参与植物多个生长发育过程。ABI3是ABA信号转导途径中的关键基因。本研究采用RT-PCR技术从花生成熟种子中克隆到ABI3的同源基因AhABI3。预测该基因开放阅读框为2313 bp,编码770个氨基酸。同源比较发现Ah ABI3与拟南芥、鹰嘴豆等的ABI3具有较高的相似性;进化分析表明AhABI3与鹰嘴豆ABI3亲缘关系最近。本研究还成功构建了AhABI3的过量表达载体pCambia2300EC-Ah ABI3,为进一步研究其功能奠定了基础。 Abscisic acid( ABA),an important plant hormone,involves in multiple facets of plant growth and development. ABI3 plays important roles in ABA signaling transduction. AhABI3 gene,homologous to ABI3 gene,was cloned from the mature seed of peanut by RT-PCR. The AhABI3 gene was predicted to contain an open reading frame of 2 313 bp which encoded 770 amino acids. The homology comparison analysis showed that AhABI3 had higher similarity with ABI3 from Arabidopsis thaliana,Cicer arietinum and so on.The phylogenetic analysis indicated that AhABI3 had the closest relationship with ABI3 from C. arietinum.The over-expression vector p Cambia 2300EC-Ah ABI3 was successfully constructed,which provided a foundation for further research of AhABI3 gene function.
