淫羊藿水煎液对小鼠不同免疫水平模型的干预作用 被引量:8

摘要 目的通过构建3个小鼠免疫系统模型,研究淫羊藿水煎液(WDE)对小鼠免疫系统的综合作用关系,从而为淫羊藿临床治疗免疫系统疾病提供理论依据。方法通过构建小鼠迟发型特异性超敏反应(DTH)模型、以碳粒廓清法检测的正常小鼠和非特异性免疫低下小鼠模型、刀豆蛋白(Con A)诱导的体外小鼠脾淋巴细胞增殖3种模型,考察淫羊藿对不同水平免疫模型的调节作用。结果 WDE能够有效降低超敏反应下的脾系数和炎症因子IL-2、IL-4水平(P<0.01);碳粒廓清法中,给药组与空白组相比WDE可使正常生理状态下小鼠的脾系数和廓清指数明显降低(P<0.05),而同样给予环磷酰胺的模型给药组与模型组相比WDE可使免疫低下小鼠的脾系数、胸腺系数、廓清指数、吞噬指数均明显升高(P<0.05);此外,WDE能显著抑制Con A诱导的脾淋巴细胞增殖反应(P<0.01),且其作用强度呈剂量依赖性。结论 WDE对DTH小鼠和正常小鼠均有免疫抑制作用,体外亦能抑制脾淋巴细胞的增殖,并且对免疫低下小鼠有显著的免疫增强和保护作用,淫羊藿水煎液可能对免疫系统有双向调节作用。 Objective Study the comprehensive effect of water decoction of Epimedium( WDE) on immune system of mice by constrcting three immune system model. This study provides the theory basis of clinical treatment for immune system diseases by using traditional Chinese medicine. Methods The author investigated the effect of WDE on different immune model by establishing three models. There are delayed type hypersensitivity( DTH) model,normal mice and immunocompromised mice model model evaluated by carbon clearance method,and multiplication of splenic lymphocyte model induced by Con A. Results The coefficient of spleen,IL-2 and IL-4 of hypersensitivity decreased significantly( P〈0. 01) after giving WDE. Detecting by Carbon clearance method,WDE decreased coefficient of spleen,expurgation index,phagocytic count of normal mice compared with blank group( P〈0. 05). However,the WDE significant increased coefficient of spleen,expurgation index,phagocytic count of immunocompromised mice compared with model group( P〈0. 05) which were both given Cy. In addition,WDE can significantly inhibit the spleen lymphocyte proliferation reaction induced by Con A,and its strength are dose dependent. Conclusion The WDE has the effect of immunosuppression on DTH in mice and normal mice,and on restrain the malignant proliferation of spleen lymphocytes in vitro,and on enhancing and protecting the immune system in immunocompromised mice obviously. So,the WDE could have two-way adjustment function of the immune system.
