微 RNA-744-5p 靶向转化生长因子β13′非编码区对食管鳞状细胞癌细胞增殖及侵袭的影响 被引量:1

摘要 miRNA-744-5p(基因编号为 MIMAT0004945)是仅在人类、仓鼠及小鼠中表达的一种 miRNA,人及小鼠序列均为5′-UGCGGGGCUAGGGCUAACAGCA-3′,而仓鼠序列末端缺少一个腺苷酸(A),具有高度保守性。目前已发现, miRNA-744-5p 在肝细胞癌、乳腺癌、胃癌中低表达,并预示预后不良[1-3];在宫颈癌及头颈部晚期肿瘤中高表达,推测可能为鳞状细胞癌(以下简称鳞癌)的肿瘤标志物之一[4-5]。TGF-β失调通路中的3个关键因子 TGF-β1、转化生长因子β受体(transforming growth factor beta receptor,TβR)Ⅰ和TβRⅡ的表达差异已被证实在食管鳞癌进展中起着重要作用,TGF-β通路失调可促进上皮-间质转化的发展,促进食管鳞癌的增殖及远处转移[6-8]。了解 TGF-β致癌通路的上游基因及具体机制,将为阻断食管鳞癌中 TGF-β致癌通路的靶向药物研制提供重要理论依据。
