荧光法测定悬浮培养藻体叶绿素含量的研究

摘要 实验室常采用荧光检测技术实现对藻的定量检测。藻细胞沉降,导致其荧光值受静置时间影响较大,静置1min时,荧光值的衰减率达△F/F0=-9.96%,无法获得准确的藻浓度。因此,本文提出一种藻体样品制备方法,培养过程中添加琼脂粉(0.08-0.10g/100mL)作为藻液悬浮稳定剂。用该方法培养的藻液其悬浮性、稳定性均有明显提高,且静置1h后,△F/F0=-4.82%≤±5%。减少了藻体沉降对测定结果的影响,确保了荧光光度法测定叶绿素含量的准确性。 Laboratory always obtain the content of algae volume by measuring the chlorophyll a fluorescence intensity. According to the settlement of algae ceils, fluorescence value are greatly influenced by static duration, when the static duration is lmin, attenuation rate of fluorescence was △F/Fo = -9. 96% and the determination of algae density is not exact. In the article, a method of algae culture preparation was established by adding agar powder( 0. 08- 0. 10g/100mL) to the culture. The suspensibility and stability of the culture was enhanced dramatically by this method, after standing 1 hour, attenuation rate of fluorescence was △F/Fo =-4. 82% ≤ ±5%. The method enhance the accuracy of chlorophyll contents measurement by chlorophyll spectrofluorimetry and reduces the effect of culture sedimentation to the measurement.
