黄芪注射液对氯化镉诱导孕鼠胎盘毒性的拮抗作用研究 被引量:6

摘要 目的:探讨黄芪注射液对氯化镉诱导孕鼠胎盘毒性的拮抗作用。方法:将45只孕鼠随机分成A、B、C3组:A组为黄芪注射液+氯化镉实验组,B组为氯化镉实验组,C组为蒸馏水对照组。干预后,孕20 d的大鼠麻醉后心脏取血,测定血清镉(Cd)、雌激素(E2)、孕激素(P)含量,然后剖宫取出胎盘,准确称重,取部分胎盘组织进行形态观察和e-NOS、AKP的平均光密度(AOD)检测,另取胎盘组织检测镉含量;每组随机取15只胎鼠准确称重,测体尾长。结果:(1)A组血清镉和胎盘镉的含量低于B组(P<0.05);血清镉与胎盘镉含量有明显相关性。(2)B组孕激素(P)、雌激素(E2)水平均明显低于C组(P<0.01),A组P、E2水平均高于B组(P<0.01)。(3)光镜下观:B组胎盘有明显的绒毛膜间质水肿,并出现纤维斑块状坏死以及淋巴细胞浸润等炎症表现;A组病理变化明显减轻。(4)B组胎盘e-NOS、AKP的AOD较C组低(P<0.01);A组胎盘e-NOS、AKP的AOD高于B组(P<0.05)。(5)B组胎盘质量、胎鼠体质量以及体尾长较均低于C组(P<0.01);A组胎盘体质量和胎鼠体质量均高于B组(P<0.05),低于C组(P<0.05)。结论:镉诱导胎盘在组织形态结构上发生一定程度损伤,黄芪注射液对氯化镉所诱导的孕鼠胎盘毒性具有拮抗作用。 Objective: To investigate the antagonistic effect of Astragalus Injection on the toxicity induced by cadmium chloride in pregnant rats..Methods: The 45 pregnant mouse were randomly divided into three groups; Astragali Mongolici injection and cadmium chloride group (Group A), cadmium chloride group (Group B) and control group (Group C). Gestational aged mouse's blood was used to test the level of serum Cd, estrogen (E2), progesterone (P), and then the placenta was extracted. The average optical density (AOD) of and e-NOS was measured. The average optical density (AKP) was measured. Results: (1) The content of cadmium and cadmi- um in serum of group A was lower than that in group B (P〈0.05). There was a significant correlation between the content of cadmium and placental cadmium. (2) group B (P), estrogen (E2) levels were significantly lower than the group C (P〈0.01), group A (P),(E2) levels were higher than the group B (P〈0.01). (3) Under the light microscope, the placenta of group B had obvious interstitial edema, and there were inflammatory manifestations such as fibrous plaque necrosis and lymphocyte infiltrationl The pathological changes of group A were significantly reduced. (4) group B of placental e-NOS, AOD of AKP was lower than the group C (P〈0.01); group A of placental e-NOS, AOD AKP was higher than the group B (P〈0.05). (5) group B of placenta weight, fetal weight and body tail length were lower than the group C (P〈0.01); group A of placental weight and fetal weight were higher than the group B (P〈0.05), lower than the group C (P〈0.05). Conclusion: Cadmium induces a certain degree of damage in the structure of the placenta, and astragalus injection has an antagonistic effect on the toxicity induced by cadmium chloride.
