摘要
北冬虫夏草(CS-4)富含虫草多糖、虫草酸、腺苷类物质等多种生物活性成分,是野生冬虫夏草最佳的替代品,试验建立了一种超声-微波联合提取法,辅助HPLC测定CS-4菌丝体冻干粉中腺苷类活性成分。结果表明:用料液比1︰300(g/m L)的蒸馏水超声提取15 min,440 W微波提取6 min,腺苷类物质的提取率高达61.24%。经HPLC检测,对菌丝体冻干粉中5种生物活性成分(胞苷、尿嘧啶、尿苷、腺苷和虫草素)进行了定性定量分析,含量分别为92.54μg/g,305.79μg/g,1.79 mg/g,785.85μg/g和695.64μg/g。
Cora'yceps militaris (CS-4) was the best substitute of wild Cordyceps militarY, mainly contained many biological active components, such as cordyccps polysacchadde, cordyceps acid, adenosine compounds. The researches had been done to establish a kind of extraction method-ultrasonic extraction method combined with microwave, which assisted HPLC method to determine the freeze-dded power of CS-4 mycelium adenosine active components. An optimum process of extraction was identified with the ultrasonic extraction by distilled water for 15 min, microwave extraction for 6 min, microwave extraction power is 400 W, with solid-liquid ratio 1 : 300 (g/mL). Under this conditions, the extraction rate of adenosine material wad up to 61.24%.Qualitative and quantitative analysis of 5 kinds active components in freeze-dried power of CS-4 mycelium had been done, detected by HPLC, and the content of cytosine, uracil, uridine, adenosine and cordycepin was 92.54 μg/g, 305.79 μg/g, 1.79 mg/g, 785.85μpg/g and 695.64 μg/g, respectively.
