摘要
以丙烯酰-β-环糊精(ACD)和硅羟基纳米氧化镍为功能单体,乙二醇二甲基丙烯酸酯(EDMA)为交联剂,运用一步键合法制备电色谱柱,并优化实验条件。通过扫描电子显微镜(SEM)、能谱(EDS)对整体柱内固定相进行表征。结果表明,固定相颗粒键合紧密,分布均匀,氧化镍含量约为33%。测定理论塔板数达8.7×104plates/m,对缬氨酸对映体的分离度达到1.62。运用该整体柱对阿替洛尔和索他洛尔的对映体进行了拆分,均达到基线分离。
Electrochromatographic column was prepared by one-step reaction, in which the acryloy - β- cyclodextrin(ACD)and nanoscale nickel oxide modified by silanol acted as functional monomers and ethylene glycol dimethacrylate served as cross-linking agent. The experimental conditions were optimized. Stationary phase in the prepared monolithic column was characterized by scanning electron microscope (SEM)and energy dispersive spectrometer (EDS). The results showed that the stationary phase particles were tightly bonded and uniformly distributed, and the content of nickel oxide was about 33%. The theoretical plate number of the column was 8.7 × 10^4plates/m, and the resolution reached 1.62 for valine enantiomers. Two kinds of chiral drugs (Atenolol and Sotalol)were separated by the prepared column, and the baseline resolutions were achieved.
