脊髓单羧酸转运蛋白-2在小鼠急性炎性痛形成中的作用

摘要 目的 探讨脊髓单羧酸转运蛋白-2(MCT-2)在小鼠急性炎性痛形成中的作用.方法 健康雄性成年昆明小鼠32只,体重20~ 25 g,采用随机数字表法分为4组(n=8):对照组(C组)、炎性痛组(IP组)、MCT-2抑制剂α-氰基-4-羟基肉桂酸(4-CIN)组(4-CIN组)和DMSO组.IP组、4-CIN组和DMSO组右足底皮下注射5%福尔马林10 μl建立炎性痛模型.模型制备前30 min,IP组、4-CIN组及DMSO组经10 s分别鞘内注射人工脑脊液、4-CIN(100 μmol)或10% DMSO,容量均为5μl.模型制备后60 min内行痛行为评分.模型制备后120 min时采用Western blot法检测脊髓MCT-2的表达.结果 与C组比较,IP组、4-CIN组及DMSO组Ⅰ相和Ⅱ相痛行为评分升高,脊髓MCT-2表达上调(P<0.01);与IP组比较,4-CIN组Ⅰ相和Ⅱ相痛行为评分降低,脊髓MCT-2表达下调(P<0.05),DMSO组上述指标差异无统计学意义(P>0.05).结论 脊髓MCT-2参与了小鼠急性炎性痛的形成. Objective To investigate the role of spinal monocarboxylate transporter-2 (MCT-2) in the development of acute inflammatory pain in mice.Methods Thirty-two male adult Kunming mice,weighing 20-25 g,were divided into 4 groups (n =8 each) using a random number table:control group (group C),inflammatory pain group (group IP),MCT-2 inhibitor α-cyano-4-hydroxy-cinnamate group (group 4-CIN),and dimethyl sulfoxide group (group D).In IP,4-CIN and DMSO groups,the inflammatory pain was induced by injection of 5% formalin 10 μl into the plantar surface of the right hindpaw.At 30 min before formalin injection,the artificial cerebrospinal fluid,4-CIN (100 μmol) and 10% DMSO 5 μl were injected intrathecally over l0 s in IP,4-CIN and DMSO groups,respectively.Pain behavior was scored within 60 min after establishment of the model.The lumbar segment of the spinal cord was harvested at 60 min after establishment of the model for detection of the expression of MCT-2 by Western blot analysis.Results Compared with group C,the pain behavior score was significantly increased in phase Ⅰ and phase Ⅱ,and the expression of MCT-2 was up-regulated in IP,4-CIN and DMSO groups (P〈0.01).Compared with group IP,the pain behavior score was significantly decreased in phase Ⅰ and phase Ⅱ,and the expression of MCT-2 was down-regulated in group 4-CIN (P〈0.05),and no significant change was found in the parameters mentioned above in group DMSO (P〉0.05).Conclusion Spinal MCT-2 is involved in the development of acute inflammatory pain in mice.
