煎煮方法对参芪养血方制剂质量的影响

摘要 目的:研究不同煎煮方法对参芪养血方制剂质量的影响,为补益类方药的制备方法选择提供依据。方法:以出膏率、正丁醇提取物和黄芪甲苷含量为评价指标。采用超高效液相色谱-四级杆串联飞行时间质谱联用法测定黄芪甲苷含量,采用ACQUITYTMC_(18)色谱柱(2.1 mm×50 mm,1.7μm),流动相乙腈-0.1%甲酸水,梯度洗脱,流速0.45 m L·min^(- 1),柱温40℃,质谱条件采用电喷雾离子化方式,在负离子模式下对黄芪甲苷进行含量测定,[M+COOH]-,m/z 829.45。结果:黄芪甲苷进样量在0.098~9.8 ng线性关系良好(r=0.999 0),平均加样回收率100.32%;参芪养血方3种制剂黄芪甲苷含量0.193~1.976 mg/剂,固含量31.297~103.077 g/剂,正丁醇提取物6.11~14.55 g/剂。结论:参芪养血方3种制剂的出膏率、正丁醇提取物含量和黄芪甲苷含量存在较大差异,制备工艺对评价指标影响较大;参芪养血方以真空浓缩机煎煮法制备为佳。 Objective: To study the effects of different decoction methods on the quality of Shenqi Yangxue preparations, and provide the evidence for choosing the preparation method of tonifying recipes. Method : Ratio of extraction, n:BuoH extract content and astragaloside Ⅳ content were used as the evaluation indexes. UPLC-Q-TOF-MS method was adopted to determine the astragaloside 1V content. ACQUITYTM C18 (2.1 mm× 50mm, 1.7μm) column was used with acetonitrile-0. 1% formic acid water as the mobile phase for gradient elution. The flow rate was set to 0.45 mL.min and the column temperature was 40 ℃. The mass spectra were obtained by Xevo G2 Q-TOF with electrospray ionization source in negative ion mode to determine the content of astragaloside IV [ M + COOH] - , m/z 829.45. Result: astragaloside Ⅳ had good linear relationship within the range of 9.8 × 10-5-9.8 × 10-3g (r = 0. 999 0) with an average recovery rate of 100.32%. The astragaloside IV contents of 3 kinds of Shenqi Yangxue preparations were 0. 193-1. 976 rag/dose, with total solid weight of 31. 297-103. 077 g/dose and n-BuoH extract content of 6. 11-14.55 g/dose. Conclusion: 3 preparation methods had significant difference in ratio of extraction, n-BuoH extract content and astragaloside IV content, so the preparation process had great effect on the evaluation indexes. The preparation method by vacuum concentrated machine had the best effect.
