摘要
目的探究大剂量盐酸异丙肾上腺素(ISO,15mg/kg)与小剂量盐酸异丙肾上腺素(3mg/kg)诱发大鼠心肌损伤模型,阐明心肌损伤程度,为本科教学提供好的模具。方法 Wistar大鼠30只,随机分为6组,正常对照组,ISO-2h(15mg/kg);ISO-3weeks(15mg/kg);ISO-2h(3mg/kg);ISO-3weeks(3mg/kg)。除正常组外,其余各组大鼠皮下多点一次性分别注射ISO 15mg/kg和3mg/kg。通过死亡率,形态学及胶原的表达比较两种剂量模型的损伤程度。结果注射15mg/kg ISO,心肌在2h即出现较大的点状坏死,3周出现明显的纤维化,大鼠死亡率亦较低;注射3mg/kg ISO,心肌在2h出现少量点状坏死,3周出现纤维增生。结论多点皮下一次性注射ISO 15mg/kg较注射ISO 3mg/kg病灶清晰,可出现明显的纤维化,为本科教学提供较好的模具。
Objective To investigate the model on large dose of Isoprenaline Hydrochloride (15 mg/kg) and small dose of Isoprenaline Hydrochloride (3mg/kg)-indueed myocardial injury. To clarify the degree of myocardial injury, and provide good mould for undergraduate teaching. Methods 30 Wistar male rats were randomly divided into 6 groups, control group (two groups), ISO-2h (15 mg/kg),ISO-3weeks (15 mg/kg),ISO-2h (3mg/kg), ISO-3weeks (3mg/kg).In addition to normal group,the other rats were injected with IS() 15 mg/kg and 3 mg/kg,respectively. The damage de- gree of the model was determined by morphology and Masson staining. Results the myocardium appeared necrosis after injection 15 mg/kg IS() in 2h and appeared the fibrosis at a weeks,the mortality of rats was lower. The myocardial tissue had a small amount of necrosis after injection ISO in 2h and appeared fiber hyperplasia. Conclusion The model of myocardial injury in injection 15 mg/kg ISO was clearer than the injection of 3 mg/kg,which could appear obvious fi brosis,could provide better mould for undergraduate teaching.
