摘要
目的:研究正常妊娠与重度子痫前期(preeclampsia,PE)患者胎盘组织中miR-221/miR-222与c-Kit的差异性表达,以探讨其对于重度子痫前期患者的意义。方法:收集2012年11月至2013年5月在深圳市妇幼保健院产科就医且行剖宫产的重度PE患者及正常妊娠胎盘组织20例,应用逆转录聚合酶链反应(RT-PCR)方法检测miR-221/miR-222差异性表达,应用逆转录聚合酶链反应(RT-PCR)方法和(Western Blot)方法检测胎盘组织中c-Kit基因的mRNA和蛋白的表达水平,探讨二者与子痫前期发病的相关性。结果:重度子痫前期胎盘中miR-221/miR-222的表达水平高于正常妊娠(P<0.05);重度子痫前期胎盘中的c-Kit基因的mRNA和蛋白的表达水平均显著降低(P<0.05,P<0.01)。结论:miR-221/miR-222与c-Kit的差异性表达可能与重度子痫前期的发病有关,二者之间的关系有待进一步研究。
Objective: To investigate differential expression of miR-221/miR-222 and c-Kit in the human placentae between severe preeclampsia and normal pregnancy; to discuss the importance of miR-221/miR-222 and c-Kit in severe PE patient. Methods: Placental tissues were obtained from 20 pregnancies complicated by severe PE and 20 normal pregnant women at the time of caesarean section who were hospitalized in Department of Obstetrics in ShenZhen Maternal and Children's Hospital between November, 2012 and May, 2013. The differential expression of miR-221/miR-222 was detected with RT-PCR. The mRNA and protein of c-Kit in human placentae were detected with RT-PCR and Western Blot. The correlationship of miR-221/miR-222 and c-Kit in PE was investgated. Results: miR-221/miR-222 in severe PE placentae were higher than that in normal pregnant placentae ( P 〈 0.05 ) ; mRNA ( P 〈 0.05 ) and protein ( P 〈 0. 01 ) of c-Kit in severe PE placentae were significantly decreased. Conclusion: The differential expression of miR-221/miR-222 and c- Kit may be related to severe PE. Further investigations are needed into the relationship between them.
