摘要
目的:研究阶段式大鼠脾阳虚证模型的复制方法及客观评价标准。方法:32只SPF级雄性SD大鼠随机分为对照组和脾阳虚证模型组(模型组),每组16只。模型组大鼠采用两段式造模方法;第1阶段,即前15 d施加饮食不节和力竭游泳刺激;第2阶段,即后7 d每日早晚2次灌服番泻叶。为质控造模过程,以临床辨证要点为基础,建立了大鼠脾气虚和脾阳虚证的评价标准,并进行客观评价。结果:与对照组比较,模型组大鼠在第1阶段出现了消瘦、食少、神疲和乏力等症状,表明大鼠处于脾气虚状态;在第2阶段,上述变化更加严重,且有畏寒、便溏等症状,提示大鼠处于脾阳虚状态。结论:本研究中采用的两段式造模方法和模型评价标准,能够成功复制大鼠脾阳虚证模型。
Objective: To study the two-phase model establishment of limited Spleen Yang deficiency in rats and its evaluation criterion. Method: 32 male SD rats in the SPF grade were randomly divided into control and model group of Spleen Yang deficiency ( model group ), each group had 16 rats. Rats in the model group were dealt with improper diet and exhausted swimming for 15 days, then hid intragastrie Senna leaf for 7 days. The model establishment was monitored, objective evaluations were carried out according to the evaluation criterion of deficiencies of Spleen Qi and Yang, which were set up based on their highlights of differentiation of symptoms and signs. Result: Rats in mode] group showed, in the 1st phase, emaciation, less food and water intakes, physical and metal fatigue, suggesting that they were in the state of spleen qi deficiency; in the 2nd phase, rats showed more severe of forementioned symptoms, adding coldness, indicating that these rats were in the state of spleen yang deficiency. Conclusion: 2-phase mode] establishment in the present study can be used to establish spleen yang deficiency in rats.
