摘要
目的观察黄芩素对人结肠癌细胞SW480移植瘤的生长及小鼠血清血管内皮生长因子(vascular endothelial growth factor,VEGF),肿瘤坏死因子-α(TNF-α)水平的影响。方法通过裸鼠前肢腋窝皮下移植的方法构建SW480细胞移植瘤模型成功后,用不同浓度的黄芩素通过灌胃给药的方式进行处理,14d后观察小鼠移植瘤的生长情况,计算肿瘤抑制率,取胸腺和脾脏计算脏器指数,并测定小鼠血清中VEGF,TNF-α的水平。结果黄芩素高、中、低剂量处理组相对于模型组,肿瘤抑制率分别为46.2%,31.3%,15.2%。高、中剂量黄芩素处理组的脏器指数显著高于模型组(P<0.05)。高剂量黄芩素处理组血清VEGF水平明显低于模型组(P<0.05),血清TNF-α水平高于模型组,差异有统计学意义(P<0.05)。结论黄芩素能抑制SW480移植瘤的生长,其可能是通过调节小鼠的免疫功能及影响VEGF的表达来发挥其抗肿瘤活性。
Objective To observe the effect of baicalein on grouth of human colonic cancer cell line SW480 in nude mice and the levels of serum VEGF and TNF-α in nude mice dealt with baicalein. Methods Transplanted colonic cancer models with SW480 cell line were established by subcutaneous injections at armpit of forelimb in nude mice. Then the nude mice were dealt with different levels of baicalein. After 14 days, we observed the growth of the tumor, calculated the growth inhibiting rate of tumor, spleen index and thymus index, and detected the levels of serum VEGF and TNF-α in nude mice. Results The growth inhibiting rates of tumor in groups dealt with high, middle and low dosage of baicalein separately were 46.2%, 31.3%, 15.2%. Organ index in groups dealt with high and middle dosage of baicalein were higher than model group(P〈0.05). In group dealt with high dosage of baicalein, the serum level of VEGF was significantly lower than model group(P〈0.05), and serum TNF-α level was higher than model group(P〈0.05). Conclusion Baicalin may inhibit the growth of SW480 cells in nude mice, and the functions could be played by regulating immune responses and influence the expression of VEGF in mice.
