摘要 建立了高效液相色谱法测定婴幼儿食品和乳品中叶黄素的方法。样品通过70℃皂化10 min,释放出叶黄素,正己烷萃取,浓缩后甲醇溶解定容,以甲醇为流动相,经C18色谱柱分离,用紫外检测器于445 nm波长处检测。回收率在96.0%~102.0%,精密度(RSD)为1.95%~3.78%,检出限分别为0.02μg/g。该方法操作简单,精密度好,准确性高,经济环保。 A simple and rapid method for determination oflutein in foods for infants and dairy products by high-performance liquid chromatography (HPLC) was developed. Following a mild saponi cation procedure (70℃ for 10 min) demonstrated to be benign to lutein, lipophilic components were partitioned by a single extraction into hexane and concentrated in mobile phase. Separation of lutein was achieved using a C18 column and isocratic elution with methanol and detected at 445 nm with a ultraviolet detector. The recovery were 96.0% - 102.0%,the relative standard deviations(RSD) were 1.95% - 3.78%, the minimum detectable values were 0.02 μg/g. This method can be used for the routine analysis oflutein.
