摘要 目的探讨CD40基因rs1883832和rs4810485位点的单核苷酸多态性(SNP)与中国辽宁地区汉族人群成人原发免疫性血小板减少症(ITP)易感相关性。方法采取病对照研究方法,选取160例成人初诊ITP住院患者(ITP组)和160例正常体检者(对照组)为研究对象。利用聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)方法测定2个SNP位点多态性分布,计算2组的基因型频率和等位基因频率。结果 rs1883832位点ITP组的CC基因型及C等位基因频率明显高于对照组(均P<0.05),TT基因型及T等位基因频率低于对照组(均P<0.05)。而rs4810485的基因型及等位基因频率2组分布差异无统计学意义(均P>0.05)。结论 CD40基因SNP位点rs1883832的C等位基因多态性可能与中国辽宁地区汉族ITP发病相关,而SNP位点rs4810485与ITP发病无相关性。 Objective To study the correlation between two single nucleotide polymorphism (rs1883832 and rs4810485 in CD40 gene ) and im- mtme thrombocytopenia (ITP) in the Liaoning region of the Han Chinese adtdt. Methods A casecontrol study was performed on 160 newly diagnosed adult 1TP patients and 160 normal controls. The polymorphism of two SNPs in CD40 gene were detected using PCR-RFLP, each genotype frequency and allele frequency was calculated. Results The CC genotype and Callele in ITP group was higher than that in control (P〈0.05). The TT genotype and T allele in ITP group was significantly lower than that in control (P 〈 0.05 ) . There was no significant difference in the genotype distri- bution or allele frequency of SNP rs4810485 in CD40 gene between the ITP and normal groups (P 〉 0.05 ). Conclusion The polymorphism of C allele CDdO gene rs1883832 may be associated with ITP in Liaoning Han Chinese, while SNP rs4810485 may not.
