高效液相色谱法同时测定大黄灵脾颗粒中丹酚酸B和淫羊藿苷的含量

Determination of Salvianolic acid B and Icariin in Dahuanglingpi Granules by HPLC

目的：建立高效液相色谱法（HPLC）用于同时测定大黄灵脾颗粒中丹酚酸 B 和淫羊藿苷的含量。方法色谱柱为 Agi-lent Zorbax SB C18（4．6 mm ×250 mm，5μm），流动相为乙腈－1．7％磷酸溶液（23∶77）等度洗脱，流速1．0 mL·min －1，柱温25℃，检测波长275 nm。结果丹酚酸 B 和淫羊藿苷分别在0．1044～6．892μg 和0．0791～2．966μg 范围内线性良好，平均加样回收率分别为102．7％和98．89％。结论建立的方法专属性强、重复性好、结果准确可靠，可用于大黄灵脾颗粒的质量控制和评价方法。

Objective To establish a HPLC method for the determination of salvianolic acid B and icariin in Dahuanglingpi granules. Methods The separation was carried on an Agilent Zorbax SB C18 column （4.6 mm ×250 mm,5 μm）and acetonitrile -1.7% phos-phoric acid solution（23 ∶77）was employed as the mobile phase.The flow rate was 1.0 mL·min -1 with UV detection wavelength at 275 nm and the column temperature was 25℃.Results The linear range of salvianolic acid B and icariin were 0.104 4 ~6.892 μg and 0.079 1 ~2.966 μg,respectively.The average recoveries were 102.7% and 98.89%.Conclusion The method is simple and accurate with good specificity and reproducibility,and it can be used for the quality control and evaluation of Dahuanglingpi granules.
