野艾蒿中两种黄酮类化合物对HepG2细胞增殖及凋亡的影响 被引量:2

摘要 目的探讨野艾蒿中提取的槲皮素和芹菜素对人肝癌Hep G2细胞增殖及凋亡的影响。方法以5-氟尿嘧啶(5-FU)为阳性对照,通过MTT法检测不同浓度槲皮素和芹菜素对Hep G2细胞的抑制作用;应用流式细胞技术检测槲皮素和芹菜素诱导Hep G2细胞凋亡的情况。结果槲皮素可抑制Hep G2细胞生长,且抑制率呈浓度依赖性关系,抑制作用强于5-FU;芹菜素也可抑制Hep G2细胞生长,其低剂量浓度对肝癌细胞无明显抑制作用,只有在高剂量浓度时抑制作用较明显,抑制作用与5-FU差异无统计学意义(P>0.05)。槲皮素和芹菜素作用于Hep G2细胞24h后,Hep G2细胞早期凋亡率和晚期凋亡率较对照组上升。结论槲皮素和芹菜素对Hep G2细胞均有明显抑制作用,其抑制机理可能是通过诱导细胞凋亡而发挥作用,可为黄酮类化合物及衍生物在抗癌方面的研究提供理论参考。 Objective To explore the effect of Quercetin and Apigenin extracted from Artemisia vulgaris on the prolifer- ation and apoptosis of HepG2 cells. Methods 5-FU was used as positive control, MTT assay was performed to test the in- hibiting effect of Quercetin and Apigenin at different concentration on HepG2 cells. The apoptosis of HepG2 cells induced by Quereetin and Apigenin was measured by flow cytometry analysis. Results Quereetin could inhibit the HepG2 cell growth with a dose dependence, and its inhibiting effect was greater than that of 5-FU; Apigenin could also inhibit the HepG2 cell growth, there was no obvious inhibitory effect at low dose concentrationon on HepG2 cell,and only at high dose concentration it was more obvious, but no significant difference in inhibiting effect was found between Apigenin and 5-FU. The early and late apoptosis rates of HepG2 cell insreased after action of Quereetin and Apigenin for 24h. Conclusion Quercetin and Apigenin can inhibit the HepG2 cell growth obviously, which may be mediated by inducing HepG2 cell apotosis. The results can provide a reference for the anticancer study of flavonoids and its derivatives.
