马尔尼菲青霉菌感染对小鼠肺泡巨噬细胞极化的影响 被引量:3

摘要 目的:研究马尔尼菲青霉菌(PM)感染诱导小鼠肺泡巨噬细胞极化的免疫反应类型。方法:以30只雄性BALB/c小鼠为实验对象,随机分为空白对照组、感染2周组、感染4周组,每组10只。麻醉处死各组小鼠后获取小鼠肺泡巨噬细胞,以每孔1×106个细胞培养24h后收集细胞上清和细胞沉淀。分别以qRT-PCR和酶活性定量方法检测精氨酸代谢关键酶及其活性的表达;以ELISA检测IL-12、TNF-α和IL-10的分泌。结果:感染2周组较空白对照组高表达经典途径激活型巨噬细胞(M1)的标志[诱导性一氧化氮合酶(iNOS)mRNA、NO、IL-12、TNF-α]和替代途径激活型巨噬细胞(M2)的标志[精氨酸酶(Arg1)mRNA、尿素](P<0.05)。以上指标在感染4周后均明显下降,感染4周组iNOS mRNA、Arg1mRNA和尿素的表达仍比空白对照组高(P<0.05);感染4周组NO、IL-12和TNF-α的表达下降至空白对照组水平或者比空白对照组水平更低。IL-10在各组的表达与空白对照组比较差异无统计学意义(P>0.05)。结论:感染PM 2周时可以明显促进小鼠肺泡巨噬细胞向M1型及可能向M2型巨噬细胞转变,在感染4周时有明显下降趋势。
