摘要
设计了一个新分子标记Wx-YMZC,改良检测Wx基因中(CT)n和G/T类型的方法,使用该标记只需要1次PCR扩增和1次酶切即可检测,简化了试验。此方法已获专利授权(专利号:ZL2013 10488947.7)。通过对55份水稻资源进行Wx基因型检测,分析和验证了不同Wx基因类型对直链淀粉含量的影响,即:1高AC(>22.0%)的资源具有较小n值(n≤14)的(CT)n重复次数的等位基因,在除糯米外的其他水稻资源中,具有低或中等AC的资源都具有较大n值(n≥16)的(CT)n重复次数的等位基因;2Wx基因中G型资源的AC高于T型资源;3Wx基因中存在着(AATT)n的重复序列,其中(AATT)6重复序列的资源多属于高AC,(AATT)5重复序列的资源多属于低或中等AC。还利用Wx-YMZC分子标记对13份R7606系列品系进行了应用研究。
Designed a new molecular marker associated with Wx gene,which had been authorized patents (ZL2013 1 0488947.7),meanwhile used this marker to detect 55 rice resources. Compared with the conventional method to detect the (CT)n and G/T type of the Wx gene, the new marker only use one PCR amplification and enzyme digestion,meanwhile, there were further analyses and validations of different Wx gene types on the influence of amylose content: 1 ) Resources of high AC had allleles with smaller numbers of (CT) repeats, (i.e.,n≤14), and conversely all resources of low to intermediate AC ,except glutinous rice resources, had alfleles with larger numbers of (CT) repeats, (i.e. ,n≥I 16);2) The amylose content with G type resources in Wx gene was higher than T ones; 3) Majorty of the resources with high AC had the (AATT)6 al- lele in the Wx gene,whereas most of the resources with low AC had the (AATT)5 allele.We analysed the (CT)n and G/T type of the Wx gene of 13 R7606 varieties.
