摘要
目的研究PI3K信号通路抑制剂2-(4-吗琳基)-8-苯基-4氢-1-苯并吡喃-4-酮(LY294002)对前列腺癌PC-3细胞增殖与凋亡的影响。方法将不同浓度的LY294002(20~40μmol/L)作用于体外培养的前列腺癌PC-3细胞24 h和48 h,用MTT比色法测定LY294002对前列腺癌细胞增殖的影响,流式细胞仪测定细胞凋亡情况,采用划痕实验及Transwell小室实验测定细胞侵袭和转移能力。结果不同浓度LY294002分别作用于PC-3细胞24 h、48 h,PC-3细胞存活率明显呈下降趋势,其中20μmol/L浓度下细胞存活率分别为(68±2)%、(45±1)%,40μmol/L浓度下细胞存活率分别为(48±2)%、(20±1)%。PC-3细胞经LY294002分别作用24 h、48 h后,细胞周期阻滞于G1期,G1期细胞比例上升,S期细胞比例下降,细胞有明显凋亡,并呈现时间和剂量依赖性(P〈0.05),其侵袭和转移能力受到抑制。结论 LY294002可显著抑制体外培养的前列腺癌PC-3细胞生长增殖,诱导其凋亡,抑制其侵袭和转移能力。
Objective To investigate the effect of PI3 K inhibitor LY294002 on the proliferation and apoptosis of prostate cancer PC- 3 cells. Methods The prostate cancer PC- 3 cells in vitro with cultured LY294002 at 20 and 40μmol / L,respectively for 24 and48 hours. The proliferation of the cells was determined by MTT assay. Cell apoptosis were measured by flow cytometry and the ability of cells invasion and metastasis were measured by Transwell and wound- healing assay. Results The prostate cancer PC- 3 cells survival rate decreased in various concentration of LY294002 for 24 and 48 hours. The cell survival rate were respectively( 68 ± 2) %,( 45 ±1) % under 40 μmol / L and( 48 ± 2) %,( 20 ± 1) % under 20 μmol / L. Meanwhile,G1 cell cycle were arrested those cells of G1 phase increased and S phase decreased in cell apoptosis,which all significantly increased in a dose and time- dependent manner( P 〈0. 05). The invasion and metastasis of PC- 3 cells were inhibited. Conclusion LY294002 can inhibit the proliferation,the ability of invasion and metastasis and induce apoptosis of prostate cancer PC- 3 cells.
