摘要
目的观察大鼠铜绿假单胞菌肺炎模型中IL-8及低氧诱导因子-1α(HIF-1α)的表达及两者之间的相关性。方法建立大鼠铜绿假单胞菌肺炎模型,ELISA法检测造模成功后3、9、24h大鼠血清中IL-8的浓度,并取3、9、24h大鼠肺组织切片行HE染色观察病理改变及免疫组化法检测HIF-1α蛋白在不同时间点的表达水平,并将IL-8及HIF-1α做相关性分析。结果在铜绿假单胞菌感染后3h大鼠血清中IL-8开始升高,9h更高,24h最高;HIF-1α的表达水平与IL-8的升高水平平行,且两者有明显相关性(r=0.974)。结论在铜绿假单胞菌肺炎的炎症过程中,IL-8与HIF-1α等炎症因子相互作用,促进炎症的发生发展。
Objective To investigate the relationship between IL-8 and HIF-1α expression in the lung tissues of rats with pseudomonas aeruginosa pneumonia model. Methods Established the model of pseudomonas aeruginosa pneumonia and detected serum IL-8 content by ELISA. Observed the pathological changes of lung tissue and the expression of HIF-1α protein by immunohistochemical staining. Analyzed the correlation of IL-8 and HIF-1α. Results In the 3 hours after infection with pseudomonas aeruginosa, the serum levels of IL-8 elevated, higher in 9 hours, the highest in 24 hours, the expression level of HIF-1α was parallel to the elevated level of IL-8, and the correlation coefficient was 0.974. Conclusion IL-8 and HIF-1α interact with each other in the inflammatory process of pseudomonas aeruginosa pneumonia, and promote the occurrence and de- velopment of pseudomonas aeruginosa pneumonia.
