摘要
目的:观察栝楼薤白半夏汤对心肌缺血再灌注损伤(MI/RI)大鼠心肌自噬及凋亡的影响。方法:将40只SD大鼠按随机数字表法分为假手术组、模型组、3-MA干预组及栝楼薤白半夏汤组,采用可逆性冠脉左前降支结扎缺血30 min、再灌注2 h复制MI/RI模型。利用全自动生化仪检测血清肌酸激酶(CK)、乳酸脱氢酶(LDH)含量,采用Western blot法测定心肌自噬基因Beclin-1、LC3II及促凋亡蛋白Bax表达水平。结果:与假手术组比较,模型组大鼠血清CK、LDH含量与心肌Beclin-1、LC3 II、Bax蛋白表达及LC3 II/LC3I水平均显著升高,与模型组比较,栝楼薤白半夏汤组及3-MA干预组能降低血清CK、LDH含量,下调心肌Beclin-1、LC3 II、Bax蛋白表达及LC3 II/LC3I水平,且栝楼薤白半夏汤抑制Beclin-1表达作用优于3-MA干预组。结论:心肌缺血再灌注可引起自噬反应增强,促进凋亡损伤,栝楼薤白半夏汤通过抑制自噬、改善心肌细胞凋亡而起到心肌保护作用。
Objective:To study the effects of Gualou Xiebai Banxia decoction(GXBD) on autophagy and antiapoptosis of myocardial ischemia-reperfusion injury(MI/RI) in rats. Methods:40 SD rats were randomly divided into sham-operated group, model group, 3-MA and GXBD groups. The model of ischemia reperfusion of the myocardium was reproduced by ligation of left descending artery for 30 min and followed by releasing the ligation for 2 hours in rats. Serum contents of CK, LDH were measured by automatic biochemical analyzer, the protein expression of Beclin-1 ,LC3II, Bax and the level of LC3 II/LC3I were determined by Western blot. Results:Compared with the sham-operated group, the contents of CK,LDH and the protein expression of Beclin-1, LC3 II, Bax and the level of LC3 II/LC3I were both significantly increased in MI/RI model groups;Compared with the model group, the 3-MA and GXBD groups down-regulated the expression of Beclin-1, LC3II, Bax and LC3 II/LC3I. The level of Beclin-1 in GXBD group were decreased than that in 3-MA group( P 〈0.05 ). Conclusion: Autophagy and apoptosis were induced and activated by MI/RI, GXBD may protect cardiomyocytes of MI/RI rats by inhibiting autophagy and reducing apoptosis.
