丙烯酸废水降解菌的筛选鉴定及降解性能研究

摘要 以汽车喷涂车间排污口污泥为菌种来源,采用丙烯酸废水为唯一碳源的筛选培养基,通过定时转接,逐步提高碳源的方法,分离、筛选出一株可降解丙烯酸废水的菌株XR002,经16S r DNA序列测序的方法对细菌进行种属鉴定及与RDPⅡ中核酸数据进行比对分析,鉴定该菌株为纤维纤维微菌。考察了温度、p H、废水质量浓度等因素对XR002菌株降解丙烯酸废水性能的影响。XR002菌株的最佳降解条件是温度为30℃、p H为9.0、丙烯酸废水初始COD质量浓度为2 000 mg/L时,7 d后丙烯酸废水的降解率可达到80%,达到了重点工业行业主要水污染物排放标准。 The research is mainly on an acrylic acid wastewater-degrading bacterium, strain XR002, filtered and separated from the outfall sludge of automotive paint shop, adopting the screening medium with the a- crylic acid wastewater as the sole carbon source, by means of timing transfer and gradual increase of the carbon source. Through the comparative analysis of 16S rDNA gene sequence and the nucleic acid data of RDP Ⅱ , the strain was identified cellulosimicrobium cellulans. It also investigates the influences on the performance of XR002 strains under the changes of pH values, temperature, concentrations of wastewater. The results show that under the optimal growth conditions for XR002 strains: within the temperature of 30℃, pH of 9.0, initial COD concentration at 2000mg/L, the absolute amount of acrylic acid wastewater degradation can reach 80% after 7 days, which can meet the regional discharge standards for main water pollutants from key industries.
