摘要 目的优化大孔吸附树脂对淫羊藿总黄酮的最佳纯化工艺。方法以淫羊藿总黄酮的洗脱率和含量为指标,通过对上样量、洗脱液浓度、洗脱液倍量等因素考察,明确淫羊藿总黄酮的纯化工艺参数。结果 D101树脂分离效果较好,优选的纯化工艺条件是以3 BV的蒸馏水洗脱,弃去水洗脱液,再以4BV的85%乙醇洗脱,收集洗脱液,流速均为1 BV/h。结论 D101型大孔树脂可用于纯化淫羊藿总黄酮,优选的纯化工艺稳定可行。 Objective To optimize purification technology of total flavonoid from Herba Epimedii with macroporous resin. Methods With elution ratio and purity of total flavonoids as indexes, optimized purification process parameters by sample volume the concentration and volume of eluent. Results D101 macroporous resin had the best separating effciency, optimized purification technology was eluting with 3BV distilled water, discarded the distilled water, eluted by the volume of 85% ethanol as eluant 4BV, collected the eluant, adsorption and desorption-power was 1 BV/h. Conclusion D101 macroporous resin could be adopted to purify total total flavonoid from Herba Epimedii, this optimized purification technology was stable and feasible.
