摘要
目的以柴胡皂苷A和柴胡皂苷D含量为指标,考察柴胡总皂苷最佳纯化方法。方法采用正丁醇萃取、D-101及AB-8大孔吸附树脂法分离纯化柴胡总皂苷,采用HPLC法测定不同纯化方法所得的总皂苷中柴胡皂苷A和柴胡皂苷D的含量。结果正丁醇萃取、D-101及AB-8大孔吸附树脂法纯化柴胡总皂苷中胡皂苷A+D的含量分别为5.93 mg/g、5.04 mg/g和10.81 mg/g,胡皂苷A+D的重量分别为26.81 mg、19.22 mg和38.70 mg。结论在本实验所用的方法中,AB-8大孔树脂法是分离精制柴胡总皂苷的最佳方法,且树脂可重复利用,具有良好的应用前景。
Objective To obtain the best purification method of Saikosaponins from Bupleuri chinese. Methods Saikosaponins were purified by n-BuOH extraction, D-101 and AB-8 macroporous resins method, and the contents of Saikosaponin A and Saikosaponin D were determined by HPLC method. Results The contents and weight of Saikosaponin A + D of Saikosaponins purified by n-BuOH extraction, D-101 and AB-8 macroporous resins method were 5.93 mg/g, 5.04 mg/g, 10.81 mg/g and 26.81 mg, 19.22 mg, 38.70 mg, respectively. Conclusion In this study, AB-8 macroporous resin purification method was the best way to separate and purificate Saikosaponins from Radix Bupleuri. As the resin could be reused, this method had a good application prospect.
