游离组织瓣缺血再灌注损伤实验动物模型的建立 被引量:5

The establishment of animal model of free flap ischemia reperfusion injury in rat

摘要 目的探讨以大鼠腹壁浅血管束为血管蒂的半侧腹壁缺血再灌注损伤动物模型的设计及应用价值。方法将30只大鼠随机分为对照组、A组(缺血3小时)和B组(缺血6小时),每组10只。制备游离组织瓣,夹闭血管蒂一段时间,随后恢复血供。评价组织瓣成活率、血流灌注、丙二醛(malondialdehyde,MDA)含量和超氧化物歧化酶(superoxide dismutase,SOD)活性。结果 A组和B组均出现不同程度的坏死,B组坏死显著。B组成活率显著低于对照组和A组(P<0.05)。B组血流灌注量明显低于对照组和A组(P<0.05);A组和B组的丙二醛含量显著升高(P<0.05),SOD活性显著下降(P<0.05)。结论以腹壁浅血管束作为血管蒂制备的大鼠腹部游离组织瓣,可作为研究组织瓣移植过程中缺血再灌注损伤的动物模型。 Objective To establish a model of ischemia reperfusion injury (IRI) of one side abdominal free flap with superficial abdomen vessel bundle in rat. Methods Thirty rats were randomly divided into three groups of ten, control group, group A and group B. The vessel bundle in the flap was clipped for 3 hours in group A and 6 hours in group B, and then the blood supply was recovered. The control group did not undergo ischemia. The flap survival rate, blood perfusion, malondialdehyde (MDA) and superoxide dismutase (SOD) in the tissue were evaluated. Results Group A and group B showed varying degree of necrosis, with the group B being more significant. The flap survival rate in group B was significantly less than that in group A and the control group ( P 〈 0. 05 ). The blood perfusion in group B was significantly lower than that in group A and the control group (P 〈 0. 05 ). The contents of MDA in group A and B significantly increased (P 〈 0. 05 ), and the activity of SOD in group A and B significantly decreased ( P 〈 0. 05 ). Conclusion The abdominal free flap with superficial abdomen vessel bundle in rat can be used as the model to study the mechanism of ischemia reperfusion injury.
